    This antibody is highly specific for Heparan Sulfate Proteoglycan 2. There is no evidence for cross-reactivity with other connective tissue proteins (vitronectin, fibronectin, elastin, collagen, laminin) but it does cross-react with human thrombospondin.

Target

  • Function
    Integral component of basement membranes. Component of the glomerular basement membrane (GBM), responsible for the fixed negative electrostatic membrane charge, and which provides a barrier which is both size- and charge-selective. It serves as an attachment substrate for cells. Plays essential roles in vascularization. Critical for normal heart development and for regulating the vascular response to injury. Also required for avascular cartilage development.
    Endorepellin in an anti-angiogenic and anti-tumor peptide that inhibits endothelial cell migration, collagen-induced endothelial tube morphogenesis and blood vessel growth in the chorioallantoic membrane. Blocks endothelial cell adhesion to fibronectin and type I collagen. Anti-tumor agent in neovascularization. Interaction with its ligand, integrin alpha2/beta1, is required for the anti-angiogenic properties. Evokes a reduction in phosphorylation of receptor tyrosine kinases via alpha2/beta1 integrin-mediated activation of the tyrosine phosphatase, PTPN6.
    The LG3 peptide has anti-angiogenic properties that require binding of calcium ions for full activity.

    Defects in HSPG2 are the cause of Schwartz-Jampel syndrome (SJS1) [MIM:255800]; a rare autosomal recessive disorder characterized by permanent myotonia (prolonged failure of muscle relaxation) and skeletal dysplasia, resulting in reduced stature, kyphoscoliosis, bowing of the diaphyses and irregular epiphyses.
    Defects in HSPG2 are the cause of dyssegmental dysplasia Silverman-Handmaker type (DDSH) [MIM:224410]. The dyssegmental dysplasias are rare, autosomal recessive skeletal dysplasias with anisospondyly and micromelia. There are two recognized types: the severe, lethal DDSH and the milder Rolland-Desbuquois form. Individuals with DDSH also have a flat face, micrognathia, cleft palate and reduced joint mobility, and frequently have an encephalocoele. The endochondral growth plate is short, the calcospherites (which are spherical calcium-phosphorus crystals produced by hypertrophic chondrocytes) are unfused, and there is mucoid degeneration of the resting cartilage.

    Proteolytic processing produces the C-terminal angiogenic peptide, endorepellin. This peptide can be further processed to produce the LG3 peptide.

No hay protocolo concreto a seguir para llevar a cabo la recuperación antigénica de los tejidos previos al uso de este anticuerpo. En nuestra web hay unos protocolos generales muy útiles, con consejos y con cada paso del protocolo detallado.

Te recomiendo que eches un vistazo al https://www.abcam.com/index.html?pageconfig=resource&rid=11384, donde hay un apartado con el protocolo recomendado para la recuperación antigénica que puede ser muy útil. Normalmente solemos recomendar recuperación antigénica con calor (olla a presión, microondas) usando tris EDTA como buffer.

Dear Sir/Madam, We are working on a laboratory located on Viet Nam and looking at Human Heparan Sulfate Measurements in dengue patient plasma. In the past, we ordered the Heparan Sulfate ELISA Kits from Seikagaku Company (Japan) to do researches related to Heparan Sulfate, these kits worked well for our experiments. However, these kits are no longer produced anymore. So, we are planning to design an ELISA protocol by ourselves for the researches in future. We found that your company has some monoclonal antibodies could be used for our experiments. We would like to ask you some information about these products. Could you please help us for the information? The products named anti –Heparan Sulfate Proteoglycan 2 antibody –A76 (ab26265) and anti-Heparan Sulfate Proteoglycan 2 antibody- A74 (ab23418) that could be used to detect for all kind of Heparan sulfate or only specific to HS Proteoglycan 2? What kind of the monoclonal antibody (ab 266265 or ab 23418) is more specific to HS if we plan to use these antibodies for coating the plates? What is the Epitope of ab 26265 antibody? It doesn’t be shown in the product instruction. The product called anti-Heparan sulfate Proteoglycan 2 antibody –A7L6 (Biotin) (ab79465) could be used for the ELISA test? This application doesn’t be suggested in the product instruction. The antibody ab79465 (Biotin) could be used as a biotinylated antibody for the ELISA plates pre- coated ab26265 or ab23418 , then it will be detected by HRP-Conjugated Streptavidin? Thank you once again for your enquiries. I was in touch with our source for these antibodies again today, and I am pleased I now have the information for you.   1. Can ab26265 and ab23418 be used to detect for all kind of Heparan sulfate or are they only specific to HS Proteoglycan 2? As far as we are aware. these antibodies are specific to HS Proteoglycan 2, but this has not been tested in the laboratory. If the epitope is in one of the domains that exists in the other heparan sulfates it might detect these,  however I am sorry the epitope has not been mapped. 2. Which is more specific to HS if we plan to use these antibodies for coating plates in ELISA? The product specification states:ab26265 is more sensitive than ab23418 in ELISA. 3. What is the Epitope of ab26265 antibody? I am sorry the epitope has not been mapped. 4. ab79465 could be used for the ELISA test? I am sorry to confirm this antibody has not been tested in ELISA.  All tested and guaranteed species and applications are listed on the online datasheet and updated as soon as we receive new information. If you would like to test this antibody in ELISA, please letme know before you purchase as you may be eligable for a testing discount. 5. I am sorry to confirm these antibodies  have not been tested together in sandwich ELISA (sELISA) and are not guaranteed for this application.  ab26265 and ab23418 have been tested in indirect or direct ELISA only. Thank you for your enquiry. Neither of the Abs that you have expressed an interest in have yet been tested for use in ICC. They have both been tested for IHC in fixed tissue sections. We do not appear to have an Ab to this target that has been specifically tested on cells. There are several resources that I would suggest: 1) Biocompare.com- a website that will allow you to search for Abs from a variety of companies. 2) The website “IHC world”: http://www.ihcworld.com/protocol_database.htm Despite the fact that this web site is focused on IHC and not ICC there is a lot of useful information that is transferable. 3) Abcam has an ICC protocol, which is accessible from the public homepage under the "protocols" hyper link, this will offer information on staining procedures e.t.c...
