Question (64186) | Anti-Hepatitis B Virus Surface Antigen antibody (HRP) (ab19990)

Question

I tried standard protocol (bicarb pH9.6 coating, blocking 1% BSA, then sandwich with your sAg standard, and detection with 2nd goat anti mouse HRP).

I ordered this Ab pair you suggested. Can you pls give me the protocol details that have worked for Abcam? I'll be using the same sAg standard that I ordered before.

Answer

Here are a few specific sELISA protocol details I was able to find:

ab2039, the capture antibody, was coated on the plate at 5 μg/ml in 0.1 M carbonate buffer, and the HRP-conjugated detection antibody, ab2042, was used at 0.5-5 ug/mL.

Otherwise, a standard sELISA protocol such as https://www.abcam.com/index.html?pageconfig=resource&rid=11422 (or your current in-house protocol) can be used with appropriate negative controls. One thing I could not find was what source for Hepatitis B Virus Surface Antigen was used in this sELISA.

I hope this is helpful. Please contact me again if you have any further questions.
