    Target

    • Relevance

      Hepatitis C E2 is a virus envelope glycoprotein which forms a heterodimer with the E1 protein. E2 inhibits human EIF2AK2/PKR activation, preventing the establishment of an antiviral state. E2 is a viral ligand for CD209/DC-SIGN and CLEC4M/DC-SIGNR, which are respectively found on dendritic cells (DCs), and on liver sinusoidal endothelial cells and macrophage-like cells of lymph node sinuses. These interactions allow capture of circulating HCV particles by these cells and subsequent transmission to permissive cells. DCs are professional antigen presenting cells, critical for host immunity by inducing specific immune responses against a broad variety of pathogens. They act as sentinels in various tissues where they entrap pathogens and convey them to local lymphoid tissue or lymph node for establishment of immunity. Capture of circulating HCV particles by these SIGN+ cells may facilitate virus infection of proximal hepatocytes and lymphocyte subpopulations and may be essential for the establishment of persistent infection.

      ELISA analysis of Hepatitis C Virus E2 binding to CD81 receptor, using ab20044 to detect Hepatitis C Virus E2 binding.

      A 96-well plate was coated with 100 ng of rho-1D4 antibody. Following overnight incubation at 4°C, the unbound antibody was washed away. The wells were blocked with blocking solution for 2 hours. 100 ng of purified CD81 was added to the wells along with E2 glycoprotein (0.4 to 20 nM) and incubated for 2 hours. ab20044 anti-Hepatitis C Virus E2 antibody (1/2000 dilution) and secondary antibody with alkaline phosphatase conjugate (1/1000 dilution) were added and incubated for 2 hours. Absorbance was measured at 495 nm by a plate-reader.

    Hello,I am looking for antibodies (preferentially mouseMAb so I can use same secondary), reacting withvirions of hepatitis A virus (HAV), hepatitis C virus (HCV) and parvovirus B19. I have selected ab64295 forB19 and ab8254 for HAV. Could you, please confirm these would detect the intactvirions of B19 and HAV? Also, I am not sure which of your anti-HCV antibodies to choose for this purpose. Could you, please advice?

    Following your enquiry I have checked all our HAV, HCV and Parvovirus B19 antibodies and doublechecked with the originators of these if the antibodies would detect intact virions. Thank you for your patience while waiting for the response.

    Concerning Parvovirus B19 antibody ab64295:
    Indeed, we can confirm that this antibody does detect the intact virus particles. This monoclonal antibody is specific for an epitope common to VP1 and VP2 structural proteins of human Parvovirus B19. These proteins do not need to be purified or the virus does not need to be disrupted in order for the antibody to recognize the target(s).

    Concerning Anti-Hepatitis A Virus antibody ab8254:
    The Hepatitis A pHM-175 strain was used as the immunogen. The clone MK-01 recognises hepatitis A virus surface antigen, unfortunately no further details are available if this antibody also detects intact virions. However, I am happy to let you know that we have ab1018 available which detects intact Hepatitis A virus:

    Concerning Anti-Hepatitis C Virus antibodies: As the envelope glycoproteins, E1 and E2, are essential components of the HCV virion, I have looked for our antibodies against these targets.
    ab54555 and ab54556: Both antibodies can detect intact Hepatitis C virions. This has so far only been tested for the inactivated form, unfortunately not yet for non-inactivated virions.
    ab20044, ab20852, ab21306 : The target protein recognized by each antibody (HCV E1 or E2) are identified by testing on Western blot. These antibodies have also been tested by ELISA and IF/IHC, but these test methods do not identify if the epitopes are recognized on intact virions. It is therefore not known if these antibodies will recognize intact HCV virions.

    I have a question regarding immunogen for ab20044. As it is stated, the immunogen was 'Recombinant full lenght E2 (genotype 1a) protein'. Could you tell me, whether the immunogen was expressed in bacterial or eukaryotic system? I would like to know, if the antibody is able to recognize glycosylated form of E2 in western blot. Also, I would like to ask, if Donkey polyclonal to Goat IgG - H&L, Cy5, ab6566, is suitable for western blot applications.

    Thank you for your enquiry. I can confrim that the immunogen for ab20044 Hepatitis C Virus E2 antibody is a recombinant antigen expressed in CHO cells. This is a polyclonal antibody and therefore recognizes multiple epitopes on the E2 protein, but we have not tested this antibody for reactivity with glycosylated E2. With regards to the secondary antibody, ab6566 is not tested in Western Blot. All tested applications are listed on the datasheets which are updated as soon as we recieve any new information.
