    HSV1 is propagated in mammalian cells using a protocol optimised for maximum particle yield. At the appropriate time, infected cells are washed in saline then disrupted to release virus particles by freeze thaw and sonication. Virus particles released from cells are partially purified by low speed centrifugation to remove cell debris. HSV1 particles are purified twice by sucrose density gradients. This process also includes a DNase step to remove cellular DNA and helps to achieve a purity of over 99%.

    ab124004 is a sucrose-purified version which is enriched for full HSV particles (including membrane and capsid). This is best when just the structural capsid and membrane proteins are desired with much less contaminating cellular material.

    Inactivation: 0.1% Triton x-100. Tested by passage on permissive cells for 5 days yielding no CPE. Vortex before use.

    This protein is derived from a pathogenic organism, and may be involved in a disease process, consequently exposure may have adverse health effects.

    Abcam has not and does not intend to apply for the REACH Authorisation of customers’ uses of products that contain European Authorisation list (Annex XIV) substances.
    It is the responsibility of our customers to check the necessity of application of REACH Authorisation, and any other relevant authorisations, for their intended uses.

    Herpes simplex type 1 (HSV-1) belongs to a family that includes HSV-2, Epstein-Barr virus (EBV) and Varicella zoster (chicken pox) virus amongst others. HSV-1 and HSV-2 are extremely difficult to distinguish from each other. Members of this family have a characteristic virion structure. The double stranded DNA genome is contained within an icosahedral capsid embedded in a proteinaceous layer (tegument) and surrounded by a lipid envelope, derived from the nuclear membrane of the last host, which is decorated with virus-specific glycoproteins spikes. These viruses are capable of entering a latent phase where the host shows no visible sign of infection and levels of infectious agent become very low.
