Target

  • Function

    Responsible for the degradation of GM2 gangliosides, and a variety of other molecules containing terminal N-acetyl hexosamines, in the brain and other tissues. The form B is active against certain oligosaccharides. The form S has no measurable activity.
  • Involvement in disease

    Defects in HEXA are the cause of GM2-gangliosidosis type 1 (GM2G1) [MIM:272800]; also known as Tay-Sachs disease. GM2-gangliosidosis is an autosomal recessive lysosomal storage disease marked by the accumulation of GM2 gangliosides in the neuronal cells. GM2G1 is characterized by GM2 gangliosides accumulation in the absence of HEXA activity, leading to neurodegeneration and, in the infantile form, death in early childhood. GM2G1 has an increased incidence among Ashkenazi Jews and French Canadians in eastern Quebec. It exists in several forms: infantile (most common and most severe), juvenile and adult (late onset).

  • Paraffin-embedded human endometrial cancer tissue stained for HEXA using ab236953 at 1/300 dilution in immunohistochemical analysis.

    After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buhher (pH 6.0). Section was blocked with 10% normal goat serum for 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using a HRP conjugated SP system.

  • PC-3 (Human prostate adenocarcinoma cell line) cells stained for HEXA (green) using ab236953 at 1/100 dilution in ICC/IF, followed by an Alexa-Fluor®488-conjugated goat anti-rabbit IgG (H+L). Counterstained with DAPI (blue).

    The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal goat serum. The cells were then incubated with primary antibody overnight at 4°C.
