- IP: HeLa whole cell lysate. IHC-P: Human ovarian carcinoma tissue.
ab245420 has not performed satisfactorily when used for WB of HIF1 beta in crude preparations (e.g. whole cell lysate). This antibody can be used for WB of enriched (e.g. immunoprecipitated) sources of HIF1 beta.
Reproducibility is key to advancing scientific discovery and accelerating scientists’ next breakthrough.
Abcam is leading the way with our range of recombinant antibodies, knockout-validated antibodies and knockout cell lines, all of which support improved reproducibility.
We are also planning to innovate the way in which we present recommended applications and species on our product datasheets, so that only applications & species that have been tested in our own labs, our suppliers or by selected trusted collaborators are covered by our Abpromise™ guarantee.
In preparation for this, we have started to update the applications & species that this product is Abpromise guaranteed for.
We are also updating the applications & species that this product has been “predicted to work with,” however this information is not covered by our Abpromise guarantee.
Applications & species from publications and Abreviews that have not been tested in our own labs or in those of our suppliers are not covered by the Abpromise guarantee.

|IHC-P||1/1000 - 1/5000. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.|
FunctionRequired for activity of the Ah (dioxin) receptor. This protein is required for the ligand-binding subunit to translocate from the cytosol to the nucleus after ligand binding. The complex then initiates transcription of genes involved in the activation of PAH procarcinogens. The heterodimer with HIF1A or EPAS1/HIF2A functions as a transcriptional regulator of the adaptive response to hypoxia.
Sequence similaritiesContains 1 basic helix-loop-helix (bHLH) domain.
Contains 1 PAC (PAS-associated C-terminal) domain.
Contains 2 PAS (PER-ARNT-SIM) domains.

HIF1 beta was immunoprecipitated from HeLa (human epithelial cell line from cervix adenocarcinoma) whole cell lysate (1 mg for IP, 20% of IP loaded) with ab245420 at 3 µg/mg lysate. Western blot was performed from the immunoprecipitate using ab245420 at 0.4 µg/ml.
Lane 1: ab245420 IP in HeLa whole cell lysate.
Lane 2: Control IgG IP in HeLa whole cell lysate.
Detection: Chemiluminescence with exposure time of 10 seconds.
Formalin-fixed, paraffin-embedded human ovarian carcinoma tissue stained for HIF1 beta using ab245420 at 1/1000 dilution in immunohistochemical analysis. Detection: DAB staining.
