ICC/IF Use at an assay dependent concentration. PubMed: 21873469
WB Use a concentration of 1 - 4 µg/ml. Detects a band of approximately 74 kDa (predicted molecular weight: 74 kDa).Can be blocked with Histidine decarboxylase peptide (ab55801).
IHC-P Use a concentration of 5 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

  • Western blot analysis of HDC expression in rat fetal liver tissue lysate.
  • IHC image of ab37291 staining in human normal skin formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab37291, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Thank you for your reply. I can see that my colleagues are assisting you with ab3611 and ab15323. The protocol suggestions that I have given you are specific for ab37291 and I see that they have also offered protocol suggestions for these products. Regarding ab37291, I would recommend decreasing the antibody concentration as we have found this to work well down to 0.5ug/ml. Thank you for contacting Abcam regarding ab37291. I am sorry that you have been experiencing difficulties with this antibody in WB. I have reviewed the protocol information you provided and I would like to make some suggestions to improve your results. Regarding the concentration of primary antibody being used, 2ug/ml is quite high. I would recommend decreasing the concentration to 1 or 0.5ug/ml which should reduce the background you observe. It may also be necessary to reduce the incubation time to 1-2 hours at room temperature rather than overnight. I hope this information is helpful.
