ELISA 1/5000.
ICC/IF 1/500 - 1/1000.

  • All lanes : Anti-Histone H1 antibody (ab61177) at 1/500 dilution

Lane 1 : extracts from COLO cells
Lane 2 : extracts from COLO cells with synthesized peptide

    Predicted band size: 36 kDa

  • IHC image of ab61177 staining Histone H1 in human colon formalin fixed paraffin embedded tissue sections, performed on a Leica Bond. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab61177, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. No primary antibody was used in the negative control (shown on the inset).

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

  • ab61177 at 1/500 - 1/1000 dilution staining Histone H1 in HuvEc cells, treated with serum (20%, 15mins) by Immunofluorescence.
  • Paraffin-embedded human colon carcinoma tissue stained for Histone H1 with ab61177 at 1/50 dilution in immunohistochemical analysis.

The sample in the right-hand panel is incubated with the immunizing peptide.
