  • Relevance

    HIV1 is equipped with the envelope gp160 glycoprotein for interaction with Langerhans cells (LCs) and dendritic cells (DCs), the members of the innate immune system, which confront the virus at the portal of virus entry in the human body. These cells are equipped with receptors by which they bind and endocytose the virus. The gp120 glycoprotein is used for binding to CD4 receptor and CCR5 co-receptor of T helper 2 (Th2) cells, and is able to induce FcepsilonRI(+) hematopoietic cells to produce IL4, which inactivates the host adaptive immune response.

    Question

    Thank you for answer and help. I will try again with more normal brain samples and with reduction of protein. I loaded 20-30 ug of brain lysate and 1.5 ul from ur recombinant protein as positive control. I will repeat again and let you know maybe we can get one more gp120 gt ab in exchange of rabbit one or I will look for any other product that we may need from Abcam. I will keep you updated. The strong band that we got with GP120 looks like specific but the problem is it appear in normal brain too.

    I have tested the protein that you sent me and I attached the results.
    Both abs that I have from Abcam binds the recombinant protein GP120 ( as two bands) but it looks like for the brain lysates rb GP120 is non specific ( got same pattern small intesity bands in cell lysates).
    Gt GP120 it recognizes a band around 60-70 kDa ( that I thought is unglycosilated GP120 )but the problem is is binds also in the normal brain lysates ( any guess or suggestions?)
    I attached the figures and if I would have a scientific input on this I will really appreciated. Regarding the antibodies, could we try more if you have any samples available?
    I have use as negative control SKN Sh cell line and normal brain lysate. All dilutions were 1:1000.

    Thank you very much for your email and for keeping me updated about the results with the recombinant protein.

    I am sorry to see that the results are still not as expected. From your images, it seems that the bands in both of the brain lysates are non-specific, since the same bands are appearing in infected and non-infected samples. The banding pattern in the brain lysates is also different using ab21179 and ab106578, which also tells me this is non-specific binding. Has the GP120 titer been measured in the infected brain sample? My only explanation is that the level of the antigen may be below the detection limit of the antibody, but I'm not sure that is the case.

    The GP120 protein ab69717 might have some degradation products or impurities at lower molecular weights as well, which appear as the bands around 40 and 55 kDa. How much of this protein was loaded per lane? We suggest 0.1-1 ug/lane, and loading less protein should decrease the visibility of these bands.

    We don't have any samples of our antibodies, but I'd be happy to send you a replacement antibody if it will be helpful for you. Which antibody would you like to try?
