  • Function

    This protein specifically binds to the DNA sequence 5'-GGGACTTTCC-3' which is found in the enhancer elements of numerous viral promoters such as those of SV40, CMV, or HIV-1. In addition, related sequences are found in the enhancer elements of a number of cellular promoters, including those of the class I MHC, interleukin-2 receptor, and interferon-beta genes. It may act in T-cell activation. Involved in activating HIV-1 gene expression. Isoform 2 and isoform 3 also bind to the IPCS (IRF1 and p53 common sequence) DNA sequence in the promoter region of interferon regulatory factor 1 and p53 genes and are involved in transcription regulation of these genes. Isoform 2 does not activate HIV-1 gene expression. Isoform 2 and isoform 3 may be involved in apoptosis.

    The molecular weight of the band on the western blot does not correspond to the molecular weight of the natural protein because only a fragment of the protein was used.

Dado que en nuestro laboratorio no se ha testado el anticuerpo con la proteína endógena, me temo que no tengo evidencias para asegurar que la banda a 34KDa corresponde a un fragmento recombinante o a un producto de degradación proteica.

Quizás aislando fracciones celulares para realizar la detección, como la fracción citoplásmica, o la nuclear, y usar un buffer de lisis como RIPA ayuden a dilucidar si realmente es un producto de degradación.

Al no tener mucha información disponible sobre la caracterización de este anticuerpo, la garantía Abpromise lo cubre si no da el resultado esperado en vuestras células.

Así pues, siendo que no hay alternativas contra HIVEP1, puedo o bien rembolsaros el importe del anticuerpo, o enviaros un reemplazo gratuito de otro anticuerpo del catálogo. También puedo emitir una nota de abono para futuros pedidos.

He confirmado con mis compañeros en el laboratorio que este anticuerpo solo se ha testado con un fragmento de la proteína recombinante, al no haberlo testado con la proteína endógena no podemos ofrecer la garantía de que el anticuerpo detecte la secuencia entera de la proteína.

Si te parece, para salir de dudas, y si estáis interesados, puedo comprobar si el péptido utilizado está disponible para que lo utilicéis para bloquear en vuestro western.

Me temo que no tenemos ningún otro anticuerpo en el catálogo para detectar HIVEP1.

The immunogen is a C-terminal fragment within residues 2550 and the C-terminus of isoform 1 - based on the UniProt database entry for the human protein P15822. http://www.uniprot.org/uniprot/P15822 The exact epitope which is usually between 5-10 residues long, has not been mapped.
