      HLA B belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta 2 microglobulin). The heavy chain is anchored in the membrane. Class I molecules play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. They are expressed in nearly all cells. The heavy chain is approximately 45 kDa. Hundreds of HLA B alleles have been described.

    Thank you for your reply and sorry for the delay in my responce.

    Unfortunately, we are not able to supply sample vials of any or our products. This is due to the size of our catalogue and the logistics this would involve. It simply isn't feasible for us.

    Additionally, as the B27 sequence and the B7 sequences are so similar, 93% homology,(SwissProt reference http://www.uniprot.org/uniprot/P03989and http://www.uniprot.org/uniprot/P01889respectively), I would not think that the antibodies we have would be likely to be able to detect the difference.

    I understand your concerns and it is regrettable the results have not been successful from this replacement vial aswell.

    As there have been difficulties with two different vials of antibody, I would appreciate if it is possible to investigate this further. This will be helpful information for our quality monitoring, and also I hope we may be able to provide some suggestions to help improve the results:

    1. Could you confirm how the samples have been prepared? Have these cells been cultured at all? Or have the cells been isolated and spun onto a slide straight away for staining (how have they been isolated?)

    2. Has a positive control been included, for example human tonsil as shown on the datasheet?

    3. How long was the fixation in PFA?

    4. Have different antigen retrieval been tried?
    As I am not sure how the samples have been prepared, so I am not currently sure if antigen retrieval is necessary. This is only usually required if the sample has been fixed in PFA for 24 hours.

    Sometimes it is possible to optimize antigen retrieval, and optimization can be different for different epitopes. For example, see the following publication for further information:

    Emoto et al., J Histochem Cytochem. 2005 53 (11) : 1311-21.

    5. Could you confirm the wash steps? We recommend 4 X for 5 minutes at each wash step in PBS containing 0.2% Tween 20. This may help to wash away any non specifically bound antibody.

    6. Is the secondary antibody working well with other primary antibodies?

    7. I can recommend to store the antibody at -20oC as soon as possible, as described on datasheet. This will ensure the antibody remains stable and we can continue to provide our guarantee. Was the antibody diluted before storage, you mention reconstitution?

    Having searched through the antibodies we have against the targets you have mentioned (HLA B7 and B27) we do not have any antibodies which are specific for only one subtype. For example, the immunogen used to raise the anti-HLA B7 antibody [EPR2623(2)], ab110645, shares high homology with multiple subtypes including B-7 and B-27. The same case is true for ab109089, although it is not expected to detect B-27. We have not tested the cross reactivity specifically and therefore cannot be sure exactly which subtype the antibodies is likely to cross react with.

    Similarly, the immunogen used to raise the anti-HLA B27 antibody, ab93150, shares 100% homology with B47, B40, B37, B44 and B13.

    In summary, we cannot guarantee that the antibodies would not cross react, in fact we would expect them to.
