    Reacts with all human classical MHC Class I molecules in native cell-surface forms. Positive in immunoblotting with many (not all) MHC Class I allelic forms (e.g. it recognizes native HLA-A2 in cytofluorometry and immunoprecipitation but not in Western blotting). Positive in Western blotting (non-reducing conditions) with most HLA-B and HLA-C molecules, but not HLA-A. Epitope specificity is very similar to classical antibody W6/32 (ab7855).

    Flow Cytometry - Anti-HLA Class I antibody [MEM-147] (ab9090)
    Overlay histogram showing Jurkat cells stained with ab9090 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab9090, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive result in 80% methanol (5 min) fixed Jurkat cells used under the same conditions.
    Please note that Abcam do not have any data for use of this antibody on non-fixed cells. We welcome any customer feedback.

    I am looking for an antibody for FACS analysis that recognizes HLA-A, B or C but not the B2 microglobulin portion of the HLA Class I antigen. I need to know if the actual HLA-A, B or C molecule is expressed on the outside of the cell and if the anitbody recognizes the B2 microglobulin part of the cell, I can't tell if just the B2 microglobulin is expressed without the HLA-A, B or C molecule or not. I am working with stem cells and the current FACS antibody I'm using recognizes the B2 microglobulin portion as well as A, B and C. So even if the cell is only expressing the B2 microglobulin, there would be a very strong postive result, but the cell does not actually express functional HLA-A, B or C. Thank you for your assistance in this matter.

Our source for ab9090 has also said that they think that a pan MHC antibody that recognizes only the free heavy chains on the surface (where it is often complexed with b2m) is not commercially available anywhere.     The supplier gave me the following information regarding your question. If you have any additional questions, please contact us again. The antibody MEM-147 (ab9090) does not react with HLA-G1 overexpressed in transfectants. It can be used for IP experiments-it recognizes most HLA-A,B,C in native conformation. In western blots-NON REDUCED NON BOILED samples only, but no HLA-A will be detected. For details see: Tran T.M. et al., Immunogenetics 53, 440 (2001) - the comparative study with W6/32 antibody.

    I'm interested in a marker for human cells - identifying cells implanted into rodent brains viewed immunohistochemically. The human cells implanted may take on any phenotype so we are not able to rely simply on something like a human specific anti-nerve specific enolase - we could get human cardiac tissue, connective tissue, blood cells, anything! What is the best choice - or range of choices for anti-human markers in such a situation? Thanks
