Tissue sections of human placental villi explants cultured in Matrigel-coated Transwells for 96 hours. Tissue was fixed with 4% paraformaldehyde, subjected to sucrose gradient for water removal, embedded in OCT medium and flash frozen. 5 ´m sections were cut in a cryostat, adhered to glass slides and allowed to air dry. Slides were washed in PBS, blocked with 10% BSA and stained overnight with 20 ´g/ml HLA-G antibody (clone 4H84, Abcam #ab52455), followed by staining with donkey-anti-mouse AlexFluor 488 secondary antibody. Nuclei were stained with DAP. Scale bars: 50 ´m.
