  • Nuclear localization of HMGB2 in PC12 cells.
    A. Cells were stained with Rabbit Anti-HMGB2 (ab67282) at 1 µg/mL

  • Cross-linking chIP was performed using HMGB2 antibody (ab67282) and 25million HUVECs. The cells were crosslinked with 15mM ethylene glycol bis(succinimidylsuccinate) (EGS) for 20 minutes at room temperature, followed by the addition of Complete Cell Fixative Solution for 40 minutes at 4°C. 4µg of HMGB2 antibody was added per 25µg of sheared chromatin. qPCR was performed using positive primers against the 5’ prime UTR OF ZNF335 and HIST2H2AC genes and negative primer against the olfactory region 8.

Answer

Thank you for your reply. I can confirm that the immunogen for ab18256 HMGB1 antibody (KPDAAKKGVVKAEKSK) has 43% alignment to the HMGB2 protein.  This is low,  so the ab18256 HMGB1 antibody is not likely to detect HMGB2. The immunogen for ab67282 HMGB2 antibody (KGKSEAGKKGPGRPTGS) has 29% alignment to the HMGB1 protein.  This is low,  so the ab67282 HMGB2 antibody is not likely to detect HMGB1. Thank you for your enquiry. The immunogen for ab67282 HMGB2 antibody has very low alignment to the HMGB1 protein, so is not likely to detect HMGB1.  However, this has not been specifically tested in the laboratory. The ab12029 HMGB1 immunogen does have higher alignment to the HMGB2 protein, and so this antibody may also detect HMGB2, but I am sorry we do not have any experimental data to confirm this.
