I agree, there is a difference between the signal you are getting with the antibody, compared to the signal that was seen when it was tested in the lab.

I still think that the antibody is detecting correct band (I have attached a copy of the image you sent, with an arrow pointing to what I think is the correct band), but there is also a high level of background. Changing the block to 5% BSA instead of milk, may help reduce the intensity of these background bands, so that the band of interest is more obvious.

If that does not prove to be successful, then we could replace the antibody with another vial of ab23392, incase the issues you are having is due to that particular vial of the antibody. Normally, I would offer to replace it with a different antibody against the same target, but we do not have any other HOXA10 antibodies in our catalogue.

You could test the antibody in nuclear vs cytoplasmic extracts for example in Western blotting to try to answer this question. You might also want to change your protocol to see if the results change or not.
