Answer

Thank you for your reply.

I agree, there is a difference between the signal you are getting with the antibody, compared to the signal that was seen when it was tested in the lab.

I still think that the antibody is detecting correct band (I have attached a copy of the image you sent, with an arrow pointing to what I think is the correct band), but there is also a high level of background. Changing the block to 5% BSA instead of milk, may help reduce the intensity of these background bands, so that the band of interest is more obvious.

If that does not prove to be successful, then we could replace the antibody with another vial of ab23392, incase the issues you are having is due to that particular vial of the antibody. Normally, I would offer to replace it with a different antibody against the same target, but we do not have any other HOXA10 antibodies in our catalogue.

Please let me know how you would like to continue.

Question
Answer

Thank you for calling Abcam earlier today.

Attached is the western blot image that we talked about on the telephone and I have highlighted the 3 bands that the antibody is detecting. Are these the same size of bands that you are seeing?

The antibody is covered under our Abpromise for six months and is guaranteed to work in western blot on human samples . If we cannot resolve the issue you are having with the antibody then I would be happy to either send a replacement antibody or to process a refund.

Please let me know if there is anything else I can help you with.

Answer

Thank you for your enquiry. We have only tested the antibody in Western blotting and so far we do not have any feedback from customers on using the antibody in IHC. You could test the antibody in nuclear vs cytoplasmic extracts for example in Western blotting to try to answer this question. You might also want to change your protocol to see if the results change or not. Our protocols are listed on the following website page: http://ops.abcam.com/index.html?pageconfig=popular_protocols Confirming the result with RNAi, knockout mouse, or a second antibody might also help. Have you tried changing the fixative, blocking conditions, or antibody dilutions or incubations? If you provide your details I can try to help you further if you like. Please fill out the form at the following link if you would like further assistance: https://www.abcam.com/index.html?section=western&pageconfig=technical&intAbID=23392&mode=questionaire I hope this information helps, please do not hesitate to contact us if you need any more advice or information.
