    HPV16 is an ATP-dependent DNA helicase required for initiation of viral DNA replication. It forms a complex with the viral E2 protein. The E1-E2 complex binds to the replication origin which contains binding sites for both proteins. HPV16, in comparison to HPV types 6 and 11, is more often associated with malignant genital cancers in humans.

My first thought was that nuclear staining should be expected, in addition to cytoplasmic, but I do not know if there is a consensus on this. I found the following article which discusses in situ hybridization data, but I do not have access to the full article:

The abstract includes this comment: "E1 and E6 transcripts were found to express with a uniquely nuclear localisation; all other transcripts had both nuclear and cytoplasmic localisation."

I see one other complaint for the antibody, which happens to also be from a researcher at Lakehead University, from 2010. Do you think you are working with the same vial of antibody? I do not see any recent orders.

Finally, I do not see any clear indication in the Wang 2004 paper that the TVG 402 clone was used for IHC, only ICC. Have you seen any data from the Doorbar lab that clearly states the 402 clone was used for IHC of sections of FFPE tissue? Here is what I found in the Methods section:

"Estimation of the number of cells showing some evidence of filament reorganization was carried out following immunostaining of HPV16-induced cervical intraepithelial neoplasia grade 1 lesions (5) using antibody TVG 405. 16E1∧E4 immunostaining of monolayer cells was carried out using either TVG 402 or TVG 405".

I am wondering if this clone in particular is incapable of detecting E1^E4 in FFPE tissues, and/or cross-reacts with an irrelevant nuclear protein. Have you tried western blotting with the TVG 402 antibody?

Thank you for the information and the reference link. I confess ignorance about the clone; I assumed our notation "E1+E4" meant reactivity with those two early antigens, not just E4. I do see that we list IHC as a tested application. My concern was that IHC was mistakenly listed, as we have no in-house IHC data for this antibody. The figure in the paper at the URL you sent confirms that it has at least stained paraffin-embedded CIN effectively.

I agree that if the originator of the clone, the Doorbar lab, has demonstrated cytoplasmic staining by IHC, then it suggests the antibody we sent in 2010 was either the wrong antibody, or a defective vial. Reviewing the record of the original complaint from the student, the staining was not primarily nuclear but rather a diffuse, non-specific stain. The more recent observation of a nuclear stain could be a consequence of further degradation of the antibody that has lead to affinity for a nuclear protein, via the variable region or some other part of the antibody, or a consequence of differences among the earlier samples and the more recent samples.

The resolution of the original complaint was a credit, issued to our distributor, Cedarlane. I am assuming that Cedarlane in turn credited the lab's account.

For our records, can you please tell me more about the tissue you are trying to stain? In particular, what is the species of the tissue, and how long was it fixed in formalin?

It is important to mention that RT-PCR data should be treated carefully since, there is no clear correlation between mRNA and protein levels and sometimes it is not easy to predict the protein expression levels from quantitative mRNA data. There are several publications available which delineate the technical boundaries of the approaches for quantitative analysis of protein expression and reveal that simple deduction from mRNA transcript analysis is insufficient.
