Target

  • Relevance

    Human Papilloma virus (HPV) is responsible for the development of type and tissue specific papilloma in the oral cavity and in the larynx. In the skin papilloma virus can cause different types of warts and in the ano-genital region it is associated with condyloma or carcinoma. HPV types 1,6 and 11 usually are associated with benign transformations of the tissues, while e.g. HPV types 16, 18 and 31 seem to be responsible for the development of some carcinoma like cervical carcinoma. The role of HPV in non-melanoic tumours of the skin is under discussion. Protein E6 is a Transcriptional transactivator, binding double stranded DNA. It has transforming activity inactivating, with E6-AP ubiquitin-protein ligase, the human TP53/p53 tumor suppressor protein by targeting it for degradation.

      Formalin-fixed, paraffin-embedded human uterine cervix squamous cell carcinoma tissue stained for HPV16 E6 + HPV18 E6 using ab70 at 1/200 dilution in immunohistochemical analysis.

      Incubated with primary antibody for 1 hour at 25°C.

      Heat mediated antigen retrieval at pH 6.

      Subconfluent proliferating MCF-7 or T47D cells were transiently transfected overnight with empty pcDNA3 vector, with FLAG wild-type E6 or zinc finger defective mutant E6 (E6-(C66,136G)). The cells were washed, post-incubated for 24 hours to allow gene expression, and harvested for Western blotting to detect the E6 oncoprotein using ab70.

    I heard back from the lab that, unfortunately, they do not have any information relating to the structure of the antibody. But this is a standard antibody so I would expect that is does have an Fc region.

    Hi,

    I would like to know the differences between the next two antibodies:

    Anti-HPV16 E6 + HPV18 E6 antibody [C1P5] (ab70) and

    Anti-HPV18 E6 antibody [289-13965] (ab30716)

    In both, the description suggest that the antibodies react with HPV 16 E6 & HPV 18 E6, however, the Specificity in antibody ab70 is not described.

    Another aspect confusing to me is the fact that the immunogen is the same in both cases: fusion protein-Gel purified HPV 18 E6- beta galactosidase fusion protein.

    Please, is really important to me understand the difference between this two options in order to choose the best for my lab.

    The antibodies ab70 and ab30716 are in fact the same antibody, produced from different sources.I can understand that this is currently not reflected very well on our website but we will be updating this. Thank you for bringing this to our attention.

    The antibodies ab70 and ab30716 are both mouse monoclonals produced from the clone C1P5 (also known as 289-13965). Both are capable of detecting both the HPV 18 E6 and HPV 16 E6, this is stated in the title of the antibody or the "specificity" note on the datasheet.

    I have located the Abreview in our system for ab70. It seems that the customer has begun this review, but must provide the product lot number (found on the vial) in order to officially submit the review. Therefore it is in an editing status.


    In general, when a review is submitted to Abcam it does not appear on the Abcam website immediately. The Scientific Support team reviews these to make sure they are complete and that the information provided makes sense. We will then publish them on the antibody datasheet.

    I sincerely apologize for the extended delay in getting this information for you, and I appreciate your patience while I have been in touch with the production lab regarding this antibody. The antibody is produced in an outside lab, and due to the nature of the antigen there is no regular QC ELISA testing done. Each new lot is tested by SDS-PAGE and an isotype test after production to ensure quality, and noapplication testing isperformed.

    About your questions and suggestions for ab70 antibody, the customer
    answers are below:
    She did the assay again, changing her protocol as suggest, however the
    unspecific staining remains.
    answers:
    1- Her cells are monolayer, and she fixed with PFA for 20 min
    2- The staining is correct, and it is strong enough at negative
    control. She did a control using only secundary antibody (FITC)- w/o
    primary antibody, and it did not stain. See attached ppt.
    3- Yes, she reduced the incubation time of primary antibody (2h),
    and even so the negative control remained stained

    Thank you for your reply.

    As we have not previously tested this antibody in ICC/IF, we could not guarantee that it would work for this application, as under our Abpromise we only guarantee it to work in WB, IP& IHC-P.

    However, I think that the customer may be seeing some specific staining using the antibody, as although there is a signal in the negative control (without HPV or BPV), the signal in the positive cells is much more intense. Although it is difficult to make a accurate comparison, as the images were taken at different magnification.

    To help reduce the background, that is being seen in the negative control, I would suggest the following:

    1 - Increase blocking to 10%BSA, for 1hr at room temperature.

    2 - Reduce the primary antibody concentration to 1/750, for 2 hrs at room temperature.

    Hopefully, the above protocol variations will help to improve the staining that is being seen.
