  • Relevance

    HRG beta 1 is an isoform from the Neuregulin/Heregulin family of structurally related polypeptide growth factors which are derived from alternatively spliced genes (NRG1, NRG2, NRG3 and NRG4). To date, there are over 14 soluble and transmembrane proteins derived from the NRG1 gene. The receptors for all NRG1 isoforms are the ERBB family of tyrosine kinase transmembrane receptors. Through interaction with ERBB receptors, NRG1 isoforms induce the growth and differentialtion of epithelial, neuronal, glial, and other types of cells. HRG1-ß1 contains an Ig domain and an EGF-like domain that is necessary for direct binding to receptor tyrosine kinases erb3 and erb4. Although HRG1-ß1 biological effects are still unclear, it has been found to promote motility and invasiveness of breast cancer cells which may also involve up-regulation of expression and function of the autocrine motility promoting factor (AMF).

  • IHC image of HRG beta 1 staining in Human normal liver formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab87547, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

I compared the homology of the sequence and Pro-neuregulin-1, membrane-bound isoforms (Q02297 NRG1_HUMAN). The only isoforms which had 100% were 6 (beta1, 71kDa) and 11 (Type IV-beta1a, 65,5kDa). 100%-isoforms 6,11 84%-isoforms 7 83%-isoforms 8,9,10 80%-isoforms 2 64%- isoforms 1,3,4 Therefore the antibody is specific for beta 1 in addition to Type IV-beta1a. We have not carried out any neutralising experiments.
