  • General notes

The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.

One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

Target

  • Relevance

Horseradish Peroxidase (HRP) is an enzyme commonly used as an indicator for chemical reactions which produce peroxide. The enzyme is routinely conjugated to antibodies for use in enzyme-based immunoassay systems. HRP functions in the removal of H2O2 (hydrogen peroxide), oxidation of toxic reductants, biosynthesis and degradation of lignin, suberization, auxin catabolism, response to environmental stresses such as wounding, pathogen attack and oxidative stress. These functions might be dependent on each isozyme/isoform in each plant tissue.
  • Cellular localization

Secreted Probable. Vacuole Probable. Note: Carboxy-terminal extension appears to target the protein to vacuoles.

Question

is this antiserum suitable for inhibiting peroxidase activity?

Asked on Aug 01 2001

Answer

The antibody has not been tested for inhibition of peroxidase. We have introduced a new scheme to allow researchers to try an antibody in an unknown application 'risk free'. If you want to place an order with our reference LD/NS.02.08.01 we can include the antibody in this scheme (No#2 below applies in this case). 1.) Abcam will refund the product if it does not work in the application and species as stated on the datasheet. 2.) If it is used in an application or on a species that is not listed on the datasheet then Abcam will refund or replace the product for another of equal value as long as Abcam receives the negative data for publishing on the datasheets to help other researchers.

Answered on Aug 02 2001

Question

I have tumours grown using transfectants stably expressing the HRP enzyme and would like to stain using immunohistochemistry. Are any of your horseradish peroxidase antibodies suitable for IHC in either frozen or paraffin embeded sections, and would they also pick up other peroxidases present in the tissue such as myeloperoxidase?

Asked on May 11 2001

Answer

We do not have any reports of any of our HRP antibodies being used as a primary antibody with frozen or paraffin embedded sections. In addition, cross-reactivity with endogenous tissue peroxidases has not been tested.
