    Muchas gracias por enviarnos el cuestionario relleno y las imágenes.

    Lamento muchísimo que el anticuerpo no os haya dado buenos resultados, y después de haber estudiado el protocolo enviado me gustaría haceros algunas sugerencias para intentar mejorarlos.

    En caso de que no se consiga obtener el resultado esperado del anticuerpo, al estar éste aun en garantía puesto que se adquirió hace menos de 6 meses, estaré encantada de mandaros un reemplazo del mismo (de un lote diferente, o bien de otro anticuerpo si lo hubiera disponible contra la misma proteína), devolveros el dinero, o haceros un vale sin caducidad para una futura compra.

    Para intentar obtener tan solo la banda correspondiente a la proteína de interés, yo sugeriría las siguientes pautas:

    -Usar buffer RIPA para lisar las células, es el que solemos usar en el laboratorio y nos da buenos resultados para detectar proteínas desnaturalizadas.

    - Cargar menos cantidad de proteína por calle. Entre 10-15ug podría ser una buena cantidad.

    - Bloquear durante un periodo mayor de tiempo puede ayudar a disminuir bandas inespecíficas. Incluso probad con 5% BSA durante 2 horas.

    -Probar diluciones mayores de ambos anticuerpos, por ejemplo 1/1500 para el primario.

    -También es importante, en casos en los que se obtienen varias bandas, llevar a cabo un control no primario. Es decir, incubar el anticuerpo secundario solo, sin primario, y comprobar que no generan ninguna banda en la membrana.

    Espero haberos ayudado.     Es difícil evaluar la procedencia de las bandas sin más información.

    Te agradecería que me mandaras los pesos moleculares en la imagen, así como información acerca del protocolo seguido.

    Te adjunto un cuestionario que se rellena en pocos minutos, y que será de gran ayuda para interpretar las bandas inespecíficas.

    Buenas tardes,
    Les escribo ya que estamos teniendouna serie de problemas con el anticuerpo ab390.
    Hemos tratado de realizar un Western Blot y optimizarlo y excepto la primera vez que dio un patrón de bandas muy inespecífico, el resto de veces proporciona tan solo una imagen con manchas inespecíficas.
    Hemos tratado de cambiar las condiciones:
    - Diferente tiempo de incubación. (1 hora, temperatura ambiente y o/n a 4ºC)
    - Lavados con TBS-tween20 0.2% y subiendo el porcentaje. (sin azida sódica)
    - Bloqueo con TBS-Tween20- BSA o leche.
    - geles "caseros" o pre-hechos.
    Hemos usado anticuerpo nuevo de la alícuota que nos proporcionan para comprobar que no se hubiera estropeado con las descongelaciones, por lo que el stock se nos ha agotado (verán que he realizado un pedido de tal producto de nuevo).
    Adjunto alguna de las imágenes.
    Podrían decirme que está sucediendo? En el datasheet no observo que haya ningún dato que pueda ayudarnos.

    Gracias por contactarnos y facilitarnos los detalles y las imágenes.

    Lamento mucho que estéis teniendo problemas con este anticuerpo. Atendiendo a las imágenes enviadas, y siendo que el vial de anticuerpo se envió en el 2010 es posible que este se haya degradado. Además como mencionas los resultados han variado desde su primer uso hasta hoy. ¿Cómo se ha almacenado el anticuerpo? ¿Ha sufrido distintos ciclos de congelación / descongelación? ¿Los buffers usados eran frescos?

    Si os interesa puedo enviaros un cuestionario que recoge los pasos principales en un ensayo de WB para tratar de entender qué ha ocurrido, pero en este caso me temo que es muy posible que se deba a la degradación del anticuerpo.

    Yo os animaría a que probarais el nuevo anticuerpo enviado, y que éste se almacene a 4C (si se va a usar todo de inmediato) o a -20C o -80C si se va a usar más a largo plazo. En este último caso se aconseja además alicuotar el anticuerpo para evitar los ciclos de congelación/descongelación.

    Si al testar este nuevo anticuerpo recibido tuvierais el mismo problema, no dudéis en contactarnos de inmediato. De cualquier forma, quedo a la espera de tu respuesta para que me indiques como deseáis proceder.

    I am looking at lux CDABE from vibrio fischeri I have used this antibody and on a western it has given me 2 bands One at app 18-20KDa and another at app 30kDa. I thought that I should expect a band at app 40kDa- is this not correct?? Any suggestions would be helpful. Thanks.
