    GST (Glutathione S-Transferase) is a 26kDa protein encoded by the parasitic helminth Schistosoma japonicum and widely used in the pGEX family of GST plasmid expression vectors as a fusion protein with foreign proteins.

    Graph of ELISA measuring levels of bound GST-tagged mosquito proteins. 5g of target protein was coated onto an ELISA plate and incubated overnight at 4°C. The plate was washed and GST-tagged mosquito proteins were incubated for an hour at RT. The proteins were washed off, GST-HRP antibody (ab3416) was diluted 1/5000 in 1%BSA in PBS and added for 30 min at RT. The plate was washed and TMB substrate was added for 20 min at RT. Stop solution was added and the O.D. of the wells read at 450 nm.

    We double checked our constructs. On the left some his/thrombin tagged proteins stained with anti-HIS. On the right the same blot stained with your anti GST (stripped first)

    These proteins contain a thrombin site but no GST! Are you 100% sure you used GST isolated from Schistosoma japonicum as antigen?? It’s much easier/cheaper to express this protein recombinant in e.coli and then inject it in mice. But if you do it like that, often it contains a thrombin site because that’s introduced by the plasmid suppliers

    Thank very much for your patience as it took us more than a month to test this antibody.

    The collaborator laboratory have confirmed that they have immunized rabbits with GST isolated from Schistosoma japonicum. The following is their response

    "we tested the antibody (by ELISA) versus various 6HIS preps and have found very little, if any reaction to 6HIS. Despite this, we absorbed the antibody versus various 6HIS preps that we have coupled to gels and then retested hoping to see any kind of change. Unfortunately, no change was observed by ELISA...this doesn't mean that there wouldn't be a change in other kinds of testing but for how we can test we are not unable to see a difference before or after absorbing.

    They have also confirmed that the antibody does not have any relation to thrombin protein so it should be binding.

    I am sorry we are unable to determine what could have gone wrong with this product. Our test show antibody is specific to the target, I guess the way left is trying a different lot. So if you are interested I can send you a new lot free of charge or another antibody against the same target.

    Question

    We are using your anti-GST antibody to detect our recombinant protein. Recently someone accidently used this antibody on a His-Trombin tagged protein (where a His-probe was supposed to be used). But he DID see a strong signal at the expected band size (of the rec. protein)!! We were wondering how this was possible (contamination, aspecific antibody, etc.). Therefore I was wondering what protein sequence was used for immunization?? I know it’s a free GST, but does this also contain a Thrombin sequence?

    Thank you for contacting us.

    The immunogen sequence does not contain thrombin, it is a GST isolated from Schistosoma japonicum species. The antibody should not be binding to His conjugated proteins.

    The band in His-thrombin might be a non specific binding which could be due to, high concentration of antibody used, damaged antibody or wrong storage of antibody. Could you let us know how the antibody was stored? Could you also repeat the experiment to check this further?

    I can confirm that ab3416 and ab106222 are different antibodies.

    I would like to reassure you that our antibodies are covered by our 6 month guarantee in the tested applications and species listed on the individual datasheets.
