    WB 1/1000 - 1/5000. Predicted molecular weight: 24 kDa. In Western blot analysis of natural cell products or Human body fluids, multiple bands of IL-6 will appear due to the variable amount of glycosylation on the molecule.

    • Function

      Cytokine with a wide variety of biological functions. It is a potent inducer of the acute phase response. Plays an essential role in the final differentiation of B-cells into Ig-secreting cells Involved in lymphocyte and monocyte differentiation. It induces myeloma and plasmacytoma growth and induces nerve cells differentiation Acts on B-cells, T-cells, hepatocytes, hematopoeitic progenitor cells and cells of the CNS. Also acts as a myokine. It is discharged into the bloodstream after muscle contraction and acts to increase the breakdown of fats and to improve insulin resistance.
    • Involvement in disease

      Genetic variations in IL6 are associated with susceptibility to rheumatoid arthritis systemic juvenile (RASJ) [MIM:604302]. An inflammatory articular disorder with systemic-onset beginning before the age of 16. It represents a subgroup of juvenile arthritis associated with severe extraarticular features and occasionally fatal complications. During active phases of the disorder, patients display a typical daily spiking fever, an evanescent macular rash, lymphadenopathy, hepatosplenomegaly, serositis, myalgia and arthritis.
      Note=A IL6 promoter polymorphism is associated with a lifetime risk of development of Kaposi sarcoma in HIV-infected men.

      Question

      I would like to perform an ELISA with IL6 specific antibodies. Since I will need to immobilize the capture antibodies myself, I need to dilute both the capture and detection antibodies. Additionally I prefer to use HRP-conjugated detection antibodies. Since the IL6 Elipair kit (ab48479) only comes with a separate HRP-Streptavidin solution, I tried to find an antibody pair and standard myself. I have selected the following antibodies/standard:

      - Anti-IL6 antibody (ab6672)
      - Anti-IL6 antibody (HRP) (ab106024)
      - IL6 standard (ab134672)

      Have these antibodies been tested to be compatible as a pair? Do you have recommended buffer solutions for:

      - Coating buffer
      - Detection buffer
      - Blocking buffer
      - Wash buffer

      I could only find a possibly suitable blocking buffer (10X Blocking Buffer (ab126587)) online. I could not find any of the other buffers sold separately.

      Answer

      Thank you for your inquiry.

      The pair of antibodies ab6672 and ab106024 may work together in sELISA, but since we haven't tested them together unfortunately we just aren't sure. Since they are both polyclonal, they will recognize multiple epitopes, but we don't know if any of those epitopes overlap.

      We do have 2 pairs of IL6 antibodies in the catalogue that have been tested in sELISA, but the corresponding detection antibodies are biotinylated. You would have to use HRP-Strep in this case. Unfortunately we don't have an HRP-conjugated IL6 that we know works well in sELISA. It sounds like you're detecting human IL6, so the second set is probably best.
