Target
-
Function
Can degrade fibrillar type I, II, and III collagens. -
Tissue specificity
Neutrophils. -
Sequence similarities
Belongs to the peptidase M10A family.
Contains 4 hemopexin-like domains. -
Domain
The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme. -
Cellular localization
Cytoplasmic granule. Secreted > extracellular space > extracellular matrix. Stored in intracellular granules. - Information by UniProt

HRP Anti-MMP8 antibody [EP1252Y] (ab206065) at 1/1000 dilution + Lung (Rat) Tissue Lysate at 10 µg
Developed using the ECL technique.
Performed under reducing conditions.
Predicted band size: 53 kDa
Observed band size: 64 kDa why is the actual band size different from the predicted?
Exposure time: 20 minutesThis blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 3% milk before being incubated with ab206065 overnight at 4°C. Antibody binding was visualised using ECL development solution ab133406.
