  • Function
    Catalyzes reversibly the conversion of cortisol to the inactive metabolite cortisone. Catalyzes reversibly the conversion of 7-ketocholesterol to 7-beta-hydroxycholesterol. In intact cells, the reaction runs only in one direction, from 7-ketocholesterol to 7-beta-hydroxycholesterol.

  • Involvement in disease
    Defects in HSD11B1 are a cause of cortisone reductase deficiency (CRD) [MIM:604931]. In CRD, activation of cortisone to cortisol does not occur, resulting in adrenocorticotropin-mediated androgen excess and a phenotype resembling polycystic ovary syndrome (PCOS).

  • Anti-HSD11B1 antibody (ab27501) at 0.3 µg/ml + Human Liver lysate (35 µg protein in RIPA buffer).

    Developed using the ECL technique.

    why is the actual band size different from the predicted?

    Primary incubation was 1 hour.
  • IHC image of ab27501 staining in human normal liver formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol B. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab27501, 10µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Thank you for your interesting enquiry. The immunogen for this antibody (ab27501) is TSYNMDRFINK, from the C Terminus of the protein sequence according to NP_005516; NP_861420. You are correct, I have had a look on the NCBI protein data base via PubMed to BLAST this sequence, and came up with no result. The only explanation I can think of is that perhaps the data base does not includes this protein. However, if you look on the ExPASy website link on the antibody datasheet, the website shows sequence homology with the human, bovine and sheep sequence. Unfortunately, I have searched for the Pig sequence to check for alignment, but could find no data. However, I hope the sequence of the immunogen given above may help you if you would like to look into this further. I hope this helps. Please do get back to me if you have any more queries.
