    Protamines substitute for histones in the chromatin of sperm during the haploid phase of spermatogenesis. They compact sperm DNA into a highly condensed, stable and inactive complex. Protamines are cross-linked by interchain disulfide bonds around the DNA-helix. Protamines are localised in the nucleus. This protein is found only in the testis and belongs to the protamine P1 family.

2. What cell fraction has been tested and how was this prepared?

3. I can suggest it would be beneficial to consider including a whole cell lysate sampleas well as the fraction, as a type of control to assess how well the procedure is working.

4. Please provide information regarding the secondary antibody. What species was it raised and and react against? Supplier and product code? It is working well with other primary antibodies?

5. The HSP1 / Protamine P1 is a very small protein, 7 kDa. Please confirm what size was the lowest size molecular weight marker used to check it has not run off the gel?
