    Abcam’s Alpha 1 acid Glycoprotein / AGP Human in vitro competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitive measurement of Alpha 1 acid Glycoprotein / AGP in plasma and serum.


    An Alpha 1 acid Glycoprotein / AGP specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently biotinylated Alpha 1 acid Glycoprotein / AGP is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Conjugate is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is inversely proportional to the amount of Alpha 1 acid Glycoprotein / AGP captured in plate.


    The entire kit may be stored at -20°C for long term storage before reconstitution - Avoid repeated freeze-thaw cycles.

Unfortunately, we do not have any Human serum available in our catalog that could be used as control and the kit ab108854 (https://www.abcam.com/ab108854) comes with Human AGP Standard but not with control serum.

We will update the graph, as it indeed confusing. When +2, 1, 0, -1 and -2is written, it means actually 102, 101, 100, 10-1, 10-2 so 100, 10, 1, 0.1 and 0.01.

1.) The standard curve example on the datasheet is plotted in log, that is why the it seems that it goes not to 4ug/ml, but actually it does. I have requested now that this should be annotated more clearly on the datasheet. Thank you for pointing this out to us.

2.) I am happy also to confirm that this is a competition ELISA assay. The protein from the standards and samples is put into the wells at the same time as the biotinylated protein from the kit. This forces the two to compete for the binding sites of the antibody on the plate. However the fluorescing reagents will only bind to the biotin on the kit protein. So the less protein in the standard and sample, the less chance it has of outcompeting the kit protein. This does result in an inverse relationship between the vivacity of the color and the amount of AGP from the standard and samples.

3.) Yes the dilution should be 1000x. We recommend doing a serial dilution to ensure thorough mixing and accurate results. First do 100x then a further 10x.
The laboratory obtained the following results typically with serum or plasma:

Plasma 1000x avg OD 0.359

Serum 1000x avg OD 0.193

I can recommend ab108854 for your customer since this kit is tested and guaranteed for plasma and serum samples. ab108852 is not.

Please find the protocol booklet for ab108854 attached.

Plasma and serum samples can usually be stored at -20C for long term storage. These samplesneed to be fully thawed before using them with the kit ab108854.
