  • Product overview

Human Apolipoprotein CIII (APOC3) in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit (ab154131) is designed for the quantitative measurement of APOC3 in plasma, serum, urine, milk, CSF, cell lysate and cell culture samples.

An Apolipoprotein CIII specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently an Apolipoprotein CIII specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Conjugate is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of Apolipoprotein CIII captured in plate.

The entire kit may be stored at -20°C for long term storage before reconstitution - Avoid repeated freeze-thaw cycles.

  • Function

Inhibits lipoprotein lipase and hepatic lipase and decreases the uptake of lymph chylomicrons by hepatic cells. This suggests that it delays the catabolism of triglyceride-rich particles.
  • Tissue specificity

Constitutes 50% of the protein fraction of VLDL and 2% of that of HDL. Synthesized predominantly in liver and to a lesser degree in intestine.
  • Involvement in disease

Defects in APOC3 may be a cause of hyperalphalipoproteinemia (HYPALIP) [MIM:143470]. Affected individuals show high levels of alpha-lipoprotein (high density lipoprotein/HDL).
  • Sequence similarities

Belongs to the apolipoprotein C3 family.
  • Post-translational
    modifications

O-linked glycan consists of Gal-GalNAc disaccharide, further modified with up to 3 sialic acid residues. O-glycosylated on Thr-94 with a core 1 or possibly core 8 glycan.
  • Cellular localization

    Secreted.

The patient whole blood was withdrawn into BD Vacutainer Glass Whole Blood Tube (BD Model 364606) containing trisodium citrate & citric acid as the anticoagulants. We use ab154131 for testing Apolipoprotein CIII in human blood plasma. We diluted our sample 4000x, and got the numbers properly in the test range.
