Human Apolipoprotein E ELISA Kit (APOE) (ab108813)

Only the assays for APOB (ab108807) and APOE (ab108813) have been tested with CSF. They are suitable for this kind of sample. The APOA1 assays have not been tested with CSF yet, but there are plans to test ab108803. Kit ab108804, the competitive ELISA for APOA1, will not be tested with CSF, and it is not known if it is suitable for assays of CSF.

Thank you for the clarification. I am not sure why the lysis buffer is failing for you, if you mean the one listed in the protocol:

Cell Lysate: Rinse cells with cold PBS and then scrape the cells
into a tube with 5 ml cold PBS with 0.5 M EDTA. Centrifuge
suspension at 1,500 rpm for 10 min at 4°C and aspirate
supernatant.

Re-suspend pellet in ice-cold Lysis Buffer (10 mM
Tris, pH8.0, 130 mM NaCl, 1% Triton X-100, protease inhibitor
cocktail).

For every 1 x 10e6 cells add approximately 100 μL of
ice-cold Lysis Buffer. Incubate on ice for 60 minutes. Centrifuge
at 13,000 rpm for 15 - 30 minutes at 4°C and collect
supernatant for assay.

That is a fairly standard procedure for collecting cell lysates, and the lysis buffer looks similar to the Thermo product. Does their protocol recommend mechanical homogenization? Do you see an insoluble pellet after spinning down the lysate made from the buffer in the Abcam protocol, compared to the pellet you see with the other buffer? If not, then I am not sure what the problem is.

Yes, the cocktail that is used by the laboratory that validates this kit is likely to be equivalent to what you have. I recommend using the inhibitor cocktail you have as recommended on the datasheet that comes with it.
