    Abcam’s BDNF Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human BDNF in serum, plasma, and cell culture supernatants.

    This assay employs an antibody specific for Human BDNF coated on a 96-well plate. Standards and samples are pipetted into the wells and BDNF present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human BDNF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of BDNF bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

    Brain derived neurotrophic factor (BDNF) is a member of the neurotrophin family of growth factors that includes NGF, NT3, and NT4. All neurotrophins have six conserved cysteine residues and share a 55% sequence identity at the amino acid level. BDNF is a potent neurotrophic factor that supports the growth and survivability of nerve and/or glial cells. BDNF has been shown to enhance the survival and differentiation of several classes of neurons in vitro, including neural crest and placode derived sensory neurons, dopaminergic neurons in the substantia nigra, basal forebrain cholinergic neurons, hippocampal neurons, and retinal ganglial cells. BDNF is expressed within peripheral ganglia and is not restricted to neuronal target fields, raising the possibility that BDNF has paracrine or even autocrine actions on neurons as well as non neuronal cells. Expression of BDNF is reduced in both Alzheimer's and Huntington disease patients. In addition, functional studies showed that age-associated changes in BDNF-mediated pathways can enhance inflammation and increase myocardial injury after myocardial infarction in the aging heart.

Detection of mouse BDNF using the BDNF Human ELISA kit

A standard curve using the standards provided, homogenate of mouse cortex, and mouse serum were read in duplicate. Two blanks, where no sample was added, were also read.

1) 60 mg of cortex was lysed in 800uL of a buffer containing 100mM Tris, 150mM NaCl, 1mM EGTA, 1mM EDTA, 1% Triton, and protease inhibitors. The lysis was done at 4C rotating on a wheel for 2 hours.

2) lysate centrifuged at 16000xg for 20 minutes collect supernatant

3) cortex supertnatant was diluted 1:4 and serum was diluted 1:5 in Diluent A for the ELISA

4) The provided protocol was followed with the standards/samples being incubated with the plate at 4C overnight (step 2 of the Assay Method).

We were pleased with the results, as we have had trouble finding a reliable ELISA for mouse BDNF.
