    Abcam’s beta 2 Microglobulin in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of beta 2 Microglobulin levels in plasma, serum, milk, saliva, urine, CSF and cell culture supernatants.

    A beta 2 Microglobulin specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently a beta 2 Microglobulin specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Conjugate is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of beta 2 Microglobulin captured in plate.

    Defects in B2M are the cause of hypercatabolic hypoproteinemia (HYCATHYP) [MIM:241600]. Affected individuals show marked reduction in serum concentrations of immunoglobulin and albumin, probably due to rapid degradation.
    Note=Beta-2-microglobulin may adopt the fibrillar configuration of amyloid in certain pathologic states. The capacity to assemble into amyloid fibrils is concentration dependent. Persistently high beta(2)-microglobulin serum levels lead to amyloidosis in patients on long-term hemodialysis.

Thank you for contacting Abcam. I have been in contact with the labs about our kit and also have some additional details regarding our other Beta-2-Microglobulin products reactivity with porcine.

For our kits:

Regardingab99977:Unfortunately, we have not tested the capture antibody against pig Beta-2-Microglobulin; however, the detection antibody is cross reactive with porcine Beta-2-Microglobulin (B2M). This does imply high homology between porcine B2M and human B2M. Therefore, there is a good possibility that the kit will successfully react with the porcine tissue sample.

It is also important to note that this kit was neither designed nor validated with lysate samples, our kits are very accommodating to many different samples types so lysate samples willlikelywork. Should you decide to test this kit with lysate samples, we recommend diluting the samples at least 5-fold with Assay DiluentBto minimize any effects of the detergents in the lysis buffer. The samples may need to be diluted further but this would need to be determined empirically.

For the original lysate, in general would want to shoot for at least 1 mg/ml total protein (preferably more) to achieve 50-500 ug/ml after dilution. Again though, this range should just be used as a starting point and the optimal concentration would need to be determined empirically.

When preparing a lysis buffer, attached are some general guidelines for doing so along with some basic tips for sample preparation. In brief, a lysis buffer must meet the following specifications:

A) has relatively low salt content (700 mM or less)
B) does not contain sodium azide
C) does not contain >0.1% SDS
D) does not contain >10 mM reducing agents (beta-mercaptoethanol or dithiothreitol)

This would include any buffers used for immunoprecipitations, including RIPA buffer.

Forab108885:We have tested porcine plasma/ serum samples with this kit. Low cross-reactivity observed so we do not suggest this kit for porcine tissue.

For our antibodies:

The Human to Porcine Identity of B2M is 72.269% overall. However there is good homology at the C terminus and in some internal regions.

Our Anti-beta 2 Microglobulin antibody [HYB332-01] (ab27155) is raised against a region with very good homology (89%). It may be found at:https://www.abcam.com/beta-2-Microglobulin-antibody-HYB332-01-ab27155.html

1. I can confirm that the kits ab108902 (Transferrin Human ELISA Kit) and ab108788 (Albumin Human ELISA kit) were especially optimised for cell culture samples and I recommend to choose them for the experiment.

3. I can confirm that c stands for competitive ELISA and s stands for sandwich ELISA. In a competitive ELISA the competitor is labeled with biotin and the amount of the bound competitor is measured. This means that a high OD indicates a low amount of the protein of interest in your sample. A low OD means that the competitor has been ousted and there is a high amount of the protein of interest in the sample.

4. I can confirm that the main difference between these two kits is the sensitivity. Both are suitable for cell culture supernatant, urine, serum, plasma.
