    Abcam’s beta 2 Microglobulin (B2M) Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human beta 2 Microglobulin in serum, plasma and cell culture supernatants.

    This assay employs an antibody specific for Human beta 2 Microglobulin coated on a 96-well plate. Standards and samples are pipetted into the wells and beta 2 Microglobulin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human beta 2 Microglobulin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of beta 2 Microglobulin bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

    Beta 2 Microglobulin non-covalently associates with the 44 kDa alpha chain to form the HLA Class I antigen complex. Human beta 2 Microglobulin associated with HLA Class I antigens are expressed on many types of cells including lymphocytes, thymocytes, monocytes, granulocytes, platelets, endothelial cells, and epithelial cells. It is absent on erythrocytes.

Cross-reactivity with monkey samples

We used Abcam's Human B2M ELISA kit to analyze monkey (cynomolgus) samples in the hope that the antibodies would cross-react. Unfortunately, it was not the case. The kit insert suggested, for human serum samples, a dilution between 5000-fold and 100 000-fold. Not knowing if the levels in monkey would be similar, we analyzed 5 different lots of monkey serum neat and diluted 100-fold, 5000-fold and 100 000-fold. All 5 lots were below the lowest standard at all dilutions including neat. We also analyzed one lot of monkey urine and got the same results. Finally, we had included one lot of Human serum that was analyzed neat and diluted100-fold and 5000-fold as a positive control. The measured concentrations were higher than the highest standard for all dilutions including neat.

Overall, the standard curve looked good and there were no problems with the assay. The results obtained with the one lot of human serum seem to indicate that the kit can quantify B2M in human serum though more extensive testing would be needed. However, it is obvious from the results that the kit cannot measure B2M in monkey serum or urine.

This kit has not been tested for cross-reactivity with mouse samples. However, since human and mouse Beta-2M differ by 30% in their primary sequence it is unlikely this kit will detect mouse Beta 2M.
