    Abcam’s Human BMP4 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human BMP4 in cell lysates and tissue lysates.

    This assay employs an antibody specific for Human BMP4 coated on a 96-well plate. Standards and samples are pipetted into the wells and BMP4 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human BMP4 antibody is added. After washing away unbound biotinylated antibody, HRP conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of BMP4 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

However, the ELISA kits is very accommodating to most body fluids and it is likely to work well with CSF samples. For the reason that secreted proteins usually at fairly low levels in CSF samples we would recommend to consider starting with a minimal dilution such as neat (no dilution) or 2-fold.

Epitope mapping has not been performed so we do not know exactly where the antibodies are binding the BMP-4 protein. All we know is that they are binding somewhere between aa 293-408 (which looks to be the entire mature protein sequence). The immunogen was a NS0-derived recombinant human BMP-4.Finally, both the capture and detection antibody are mouse monoclonal.

Thank you for contacting us. Both kits ab99982 and ab99983 use the same antibody pairs. The homology between mouse and human BMP4 is 98% and it is therefore likely that these kits will detect mouse BMP4 as well. This has not been tested in house however, and results on mouse samples would not be guaranteed. I hope this helps, please let me know if you need any additional information.
