    CCL16 in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of CCL16 protein in human serum, plasma, and cell culture supernatants.


    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB Development Solution is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.


    CCL16 (C-C motif chemokine ligand 16) is a chemoattractant cytokine produced in the thymus, liver, and spleen. This protein is up-regulated in response to IL-10 and recruits lymphocytes and monocytes. As such, elevated levels are found during inflammatory events such as; preeclampsia, irritable bowel syndrome, and pneumonia.

  • Function

    Shows chemotactic activity for lymphocytes and monocytes but not neutrophils. Also shows potent myelosuppressive activity, suppresses proliferation of myeloid progenitor cells. Recombinant SCYA16 shows chemotactic activity for monocytes and THP-1 monocytes, but not for resting lymphocytes and neutrophils. Induces a calcium flux in THP-1 cells that were desensitized by prior expression to RANTES.

    Mainly expressed in liver, also found in spleen and thymus. Highly expressed in LPS- and IFN-gamma-activated monocytes, weakly in some lymphocytes, including natural killer cells, gamma-delta T-cells, and some T-cell clones.

  • The CCL16 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

  • The concentrations of CCL16 were measured in duplicates, interpolated from the CCL16 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1%, plasma (citrate) 1.5%, and HepG2 supernatant 30%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CCL16 concentration was determined to be 15284.9 pg/mL in serum, 12472.9 pg/mL in plasma (citrate) and 599.3 pg/mL in HepG2 supernatant.
