    CD276 in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of CD276 protein in human serum, plasma, supernatant and urine.

    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

    CD276 (B7-H3) is a glycoprotein member of the B7 family, Ig superfamily of proteins. CD276 is involved in the regulation of T-cell-mediated immune response and is expressed by a variety of cell types.

    May participate in the regulation of T-cell-mediated immune response. May play a protective role in tumor cells by inhibiting natural-killer mediated cell lysis as well as a role of marker for detection of neuroblastoma cells. May be involved in the development of acute and chronic transplant rejection and in the regulation of lymphocytic activity at mucosal surfaces. Could also play a key role in providing the placenta and fetus with a suitable immunological environment throughout pregnancy. Both isoform 1 and isoform 2 appear to be redundant in their ability to modulate CD4 T-cell responses. Isoform 2 is shown to enhance the induction of cytotoxic T-cells and selectively stimulates interferon gamma production in the presence of T-cell receptor signaling.

  • Interpolated concentrations of CD276 in human serum and plasma samples. The concentrations of CD276 were measured in duplicates, interpolated from the CD276 standard curves and corrected for sample dilution. Undiluted samples: serum 50%, plasma (EDTA) 25%, plasma (citrate) 25%, plasma (heparin) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD276 concentration was determined to be 31.7 ng/mL in serum, 25.0 ng/mL in plasma (EDTA), 25.1 ng/mL in plasma (citrate) and 28.5 ng/mL in plasma (heparin).
