    Abcam’s CD44 Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of soluble CD44 in Human serum, plasma, buffered solutions and cell culture medium.

    A monoclonal antibody specific for CD44 has been coated onto the wells of the microtiter strips provided. Samples, including standards of known CD44 concentrations, control specimens or unknowns are pipetted into these wells. During the first incubation, the standards or samples and a biotinylated monoclonal antibody specific for CD44 are simultaneously incubated. After washing, the enzyme Streptavidin-HRP, that binds the biotinylated antibody is added, incubated and washed. A TMB substrate solution is added which acts on the bound enzyme to induce a colored reaction product. The intensity of this colored product is directly proportional to the concentration of CD44 present in the samples. 

    This kit will recognize both endogenous and recombinant Human CD44.

    Receptor for hyaluronic acid (HA). Mediates cell-cell and cell-matrix interactions through its affinity for HA, and possibly also through its affinity for other ligands such as osteopontin, collagens, and matrix metalloproteinases (MMPs). Adhesion with HA plays an important role in cell migration, tumor growth and progression. Also involved in lymphocyte activation, recirculation and homing, and in hematopoiesis. Altered expression or dysfunction causes numerous pathogenic phenotypes. Great protein heterogeneity due to numerous alternative splicing and post-translational modification events.

    Isoform 10 (epithelial isoform) is expressed by cells of epithelium and highly expressed by carcinomas. Expression is repressed in neuroblastoma cells.

Thank you for your interest in the CD44 and CD56 ELISA kits, ab45912 and ab119587. Neither has been tested for reactivity with mouse or rat samples. We are unable to assess the probability of cross-reaction by BLAST, as the locations of the epitopes that the antibodies recognize are unknown. In general, without data showing otherwise, it is best to assume there will be some cross-reaction.

The laboratory confirmed thatboth antibodies will recognize the part after the cleavage site - the part which will be detached from the cells - please see here the comment from the laboratory for more details:

"The answer to the question is : the both antibodies recognize the protein AFTER the cleavage site.
I have explained with more detailed in the document added.
In a solution without cells, only the form soluble is detected because the both antibodies interacted in those soluble part. In a solution containingalso cells (culture for example), the soluble form is detected and also the membrane form could (we never have tested that but that is sure that as least one of those antibodies: x is able to recognizes the membrane form because it is also used in cytometry).
If the epitopes had been before the cleavage site, the ELISA on free cells solution would have given negative results"

Unfortunately the antibody used as detection antibody has not been tested in any other application as IHC or ICC. We do therefore not know whether it can detect the CD44 on the cells.

The coated antibody is able to recognize the cell surface protein.

If the customer wants to perform Immunocytochemistry )ICC= of Flow cytometry, I would recommend to us another antibody and not the antibodies from the kit. I can recommend for example therefore the ab119863 , the FITC conjugated ab19622 or the ab51037. These antibodies have all been tested in ICC, IHC and flow cytometry.
