    Human CYR61 ELISA Kit (ab238267) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of CYR61 protein in cell culture extracts, cell culture supernatant, cit plasma, edta plasma, milk, saliva, serum, tissue extracts, and urine. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human CYR61 with 5 pg/ml sensitivity.


    SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:


            - Single-wash protocol reduces assay time to 90 minutes or less
            - High sensitivity, specificity and reproducibility from superior antibodies
            - Fully validated in biological samples
            - 96-wells plate breakable into 12 x 8 wells strips


    A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

    CYR61 is a secreted, cysteine rich, heparin binding protein encoded by a growth factor inducible immediate early gene. It promotes cell proliferation, chemotaxis, angiogenesis and cell adhesion, and appears to play a role in wound healing by up regulating, in skin fibroblasts, the expression of a number of genes involved in angiogenesis, inflammation and matrix remodeling including VEGA A, VEGA C, MMP1, MMP3, TIMP1, uPA, PAI 1 and integrins alpha 3 and alpha 5. Cyr61 mediated gene regulation is dependent on heparin binding. CYR61 down regulates the expression of alpha 1 and alpha 2 subunits of collagen type 1. It promotes cell adhesion and adhesive signaling through integrin alpha 6/beta 1, cell migration through integrin alpha v/beta 5 and cell proliferation through integrin alpha v/beta 3.

      The concentrations of CYR61 were measured in duplicates, interpolated from the CYR61 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 100%, plasma (citrate) 100%, and plasma (EDTA) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CYR61 concentration was determined to be 182.2 pg/mL in neat serum, 123.5 pg/mL in neat plasma (citrate), and 155.3 pg/mL in neat plasma (EDTA).
