    CYR61 in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of CYR61 protein in human serum, plasma, milk, urine, saliva, cell culture supernatants, and cell and tissue extracts.


    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB Development Solution is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.


    CYR61 (Cysteine-rich Angiogenic Inducer 61), also known as CCN Family Member 1(CCN1) or Insulin-Like Growth Factor-Binding Protein 10 (IGFBP10) is a secreted protein that promotes cell proliferation, chemotaxis, angiogenesis and cell adhesion. CYR61 exhibits both tumorigenic and tumor suppressor properties, depending in part on the localization.

    CYR61 is a secreted, cysteine rich, heparin binding protein encoded by a growth factor inducible immediate early gene. It promotes cell proliferation, chemotaxis, angiogenesis and cell adhesion, and appears to play a role in wound healing by up regulating, in skin fibroblasts, the expression of a number of genes involved in angiogenesis, inflammation and matrix remodeling including VEGA A, VEGA C, MMP1, MMP3, TIMP1, uPA, PAI 1 and integrins alpha 3 and alpha 5. Cyr61 mediated gene regulation is dependent on heparin binding. CYR61 down regulates the expression of alpha 1 and alpha 2 subunits of collagen type 1. It promotes cell adhesion and adhesive signaling through integrin alpha 6/beta 1, cell migration through integrin alpha v/beta 5 and cell proliferation through integrin alpha v/beta 3.
