  • Product overview

Abcam’s Human Cystatin C in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of Human Cystatin C in cell culture supernatants, serum, plasma (heparin, EDTA) saliva, urine and Human milk.

A Cystatin C specific mouse monoclonal antibody has been precoated onto 96-well plates. Standards and test samples are added to the wells and incubated. A biotinylated detection polyclonal antibody from goat, specific for Cystatin C is then added followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. TMB is then used to visualize the HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the Human Cystatin C amount of sample captured in plate.

  • Research areas

  • Function

As an inhibitor of cysteine proteinases, this protein is thought to serve an important physiological role as a local regulator of this enzyme activity.
  • Tissue specificity

Expressed in submandibular and sublingual saliva but not in parotid saliva (at protein level). Expressed in various body fluids, such as the cerebrospinal fluid and plasma. Expressed in highest levels in the epididymis, vas deferens, brain, thymus, and ovary and the lowest in the submandibular gland.
  • Involvement in disease

Defects in CST3 are the cause of amyloidosis type 6 (AMYL6) [MIM:105150]; also known as hereditary cerebral hemorrhage with amyloidosis (HCHWA), cerebral amyloid angiopathy (CAA) or cerebroarterial amyloidosis Icelandic type. AMYL6 is a hereditary generalized amyloidosis due to cystatin C amyloid deposition. Cystatin C amyloid accumulates in the walls of arteries, arterioles, and sometimes capillaries and veins of the brain, and in various organs including lymphoid tissue, spleen, salivary glands, and seminal vesicles. Amyloid deposition in the cerebral vessels results in cerebral amyloid angiopathy, cerebral hemorrhage and premature stroke. Cystatin C levels in the cerebrospinal fluid are abnormally low.
Genetic variations in CST3 are associated with age-related macular degeneration type 11 (ARMD11) [MIM:611953]. ARMD is a multifactorial eye disease and the most common cause of irreversible vision loss in the developed world. In most patients, the disease is manifest as ophthalmoscopically visible yellowish accumulations of protein and lipid that lie beneath the retinal pigment epithelium and within an elastin-containing structure known as Bruch membrane.
  • Sequence similarities

    Belongs to the cystatin family.
  • Post-translational
    modifications

The Thr-25 variant is O-glycosylated with a core 1 or possibly core 8 glycan. The signal peptide of the O-glycosylated Thr-25 variant is cleaved between Ala-20 and Val-21.
  • Cellular localization

    Secreted.
  • Information by UniProt

Question
Answer

Urine samples should be centrifugred at 10,000 x g at 4C for 2 minutes. The supernatant can either be used immediately or aliquoted and stored at -20 or -80C. We have not performed a stability study over time to determine specifically how long the Cystatin C will be stable under these conditions.

Answer

Thank you for contacting us.
It is very important for this assay to dilute samples so that the measurements are in the range of the standard curve.
The datasheet indicates a range from 4.1 pg/ml - 1000 pg/ml (= 0.041- 1 ng/ml) for the NGAL kit. The measurement from the pathology department shows that the sample concentration is 6-500 ng/ml, which would be too high to measure undiluted with the ab113326.
For the Cystatin kit the range of the standard curve is 0.312 ng/ml - 20 ng/ml, the sample contains 1-200 ng/ml measured by the pathology department. We would recommend to dilute this sample 1:10 and also to optimise the experiment by trying different dilutions for the measurement.
Another issue could be the pH of the undiluted urine samples. A low pH of the undiluted sample might interfere with the ELISA kit. However, we would not expect this as a problem using a diluted sample.
I hope this information is helpful to you. Please do not hesitate to contact us again if no satisfactory results can be achieved with these products in the future.

Answer

Thank you for taking the time to complete our questionnaire and contact us. I am sorry to hear you have had difficulty obtaining satisfactory results from these kits.
The details you have kindly provided will enable us to investigate this case for you and this is also helpful in our records for monitoring of quality.
I would like to reassure you that both kits are tested and covered by our 6 month guarantee for use in human urine samples. In the event that a product is not functioning in the applications cited on the product data sheet, we will be pleased to provide a credit note or free of charge replacement.
I would also appreciate if you can confirm some further details:
1. Could you kindly provide the resulted standard curves and the results of the positive control for this two kits?
2. Could you please provide more information about the protein that was detected in the pathology department using the other kits? What level of protein was detcted in the used sample?
3. Could you please confirm how the samples were treated/stored prior to the ELISA? How many tests were already performed with these kits?
Thank you in advance for your cooperation. Please do not hesitate to contact me again with the further requested details.

Question

As I’m not familiar with these items, I tried to find the ab codes through your site.
For the items I found a relative code I have update the list.
For those I don’t have a reference code, this is because either no products matches or all available products reacts for humans when we need them for pigs.
So please check the list again and advice.

I have received information regarding

ab119589 Cystatin C Human ELISA Kit
ab119590 Cystatin C Mouse ELISA Kit
ab119596 KIM1 Mouse ELISA Kit
ab119597 KIM1 Rat ELISA Kit
ab119601 Lipocalin-2 / NGAL Mouse ELISA Kit
ab119601 Lipocalin-2 / NGAL Mouse ELISA Kit

these has not been tested with Swine or pig samples so we are unable to confirm the species cross reactivity. It mean only ab100755, ab100756, ab108674, ab108794, ab113352, ab113353 are OK to use with Porcine samples and are guaranteed. Would you like to have a quote for these antibodies.

I hope this info will be helpful. Should you require more information please do not hesitate to contact me.
