    Abcam’s Cytochrome c Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for accurate quantitative measurement of Human Cytochrome c concentrations in cell lysate.

    Cytochrome c specific antibodies have been precoated onto 96-well plates. Standards and test samples are added to the wells along with a biotinylated Cytochrome c detection antibody and the microplate is then incubated at room temperature. Following washing with wash buffer a Streptavidin-HRP conjugate is added to each well, incubated at room temperature and unbound conjugates are then washed away using was buffer. TMB is then added and catalyzed by HRP to produce a blue color product that changes to yellow after addition of an acidic stop solution. The density of yellow coloration is directly proportional to the amount of Cytochrome c captured in plate.

The lysis buffer in kits ab119507 and ab119521 are identical, so you can analyze lysates prepared for ab119507 with ab119521.
For ab119521, the lysis buffer allows for lysis of the cellular membrane but not other compartments with in the cytoplasm. The mitochondria remain intact, thus the kit detects only released and not mitochondrial Cytochrome C.

Thank you for your email and your interest in our products.

Unfortunately, we would not recommend using the Cytochrome c Human ELISA Kit (with color giving dyes), ab119521 with serum sample as when we have tried this we have observed false positive results. Additionally the spike recovery and dilution was not very good which indicates that serum interferes with the kit and would not give reliable results using these samples.
