    Abcam’s anti-Cytomegalovirus (CMV) IgG Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the qualitative measurement of IgG class antibodies against Cytomegalovirus in Human serum and plasma.

     

    A 96-well plate has been precoated with Cytomegalovirus antigens to bind cognate antibodies. Controls or test samples are added to the wells and incubated. Following washing, a horseradish peroxidase (HRP) labelled anti-Human IgG conjugate is added to the wells, which binds to the immobilized Cytomegalovirus-specific antibodies. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The density of yellow coloration is directly proportional to the amount of Cytomegalovirus IgG sample captured in plate.

Quantification of IgG anti-CMV ELISA in frozen plasma samples with ab108724 Anti-cytomegalovirus (CMV) IgG human ELISA kit

Quantification of human IgG anti-CMV in EDTA plasma samples (freshly frozen and stored at -80C) was tested using the ab108724 ELISA kit. The information provided with the kit stated that only serum samples and citrate plasma samples had been previously tested. We wanted to test whether the kit allowed accurate quantification of human IgG anti-CMV in EDTA plasma.
Plasma samples were diluted 1:100 with the sample diluent provided with the kit (as per the manufacturer’s instructions) and assayed in duplicate. Duplicate wells for positive control, cut-off and negative control as well as blank were included as per the protocol provided. All steps followed the instructions provided with the ELISA kit.
Absorbance (optical density, OD) was determined with a plate reader set at 450 nm with a reference wavelength of 650 nm. Absorbance of blank wells was averaged and subtracted from all other wells. Absorbance values were converted into standard units as instructed by the manufacturer (cut-off = 10 units positive > 11 units negative < 9 units grey zone = 9-11 units).
The kit distinguished clearly between IgG anti-CMV positive and negative samples and allowed quantification of these antibodies in EDTA plasma samples (see Figure).
The protocol provided with the kit was clear and easy to follow and contained all the information necessary to perform the assay.

A1: The CMV ELISA, ab108724, is qualitative and therefore does not have a known detection limit. The assay sensitivity is defined as the probability of the assay scoring positive in the presence of specific analyte. The assay sensitivity for this kit is 98%. A2: The cut-off control is used to determine the optical density threshold at which samples are considered positive or negative. Please see the product insert for details on this calculation. A3: The controls in this kit come in liquid form ready to use.

Thank you for your patience. The laboratory that developed the ELISA has sent the following information. If it does not completely answer your questions, please contact me. The CMV IgG ELISA is a qualitative test. Therefore we do not have data about the detection limit or spiking. Heat inactivation of samples is not recommended since it influences the test results in a negative way. Cross reactivity with antibodies against HSV1-2, Measles, Mumps, or VZV have not been observed. The diagnostics specificity is defined as the probability of the assay scoring negative in the absence of specific analyte. For this, it is 97.5%. The diagnostic senstivity is defined as the probability of the assay scoring positive in the presence of specific analyte. The sensitivity of this assay is 98%. Interferences with hemolytic, lipemic, or icteric sera are not ovserved up to a concentration of 10 mg/ml hemoglobin, 5 mg/ml triglycerides, and 0.2 mg/ml bilirubin.
