    • Product overview

Dengue virus ELISA kit - IgG is designed for the accurate qualitative measurement of IgM class antibodies against Dengue virus in Human serum and plasma.

A 96-well plate has been precoated with Dengue virus antigens to bind cognate antibodies. Controls or test samples are added to the wells and incubated. Following washing, a horseradish peroxidase (HRP) labelled anti-Human IgG conjugate is added to the wells, which binds to the immobilized Dengue virus-specific antibodies. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The density of yellow coloration is directly proportional to the amount of Dengue virus IgG sample captured in plate.

A horseradish peroxidase (HRP) labelled anti-Human Ig conjugate is added to the wells. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The intensity of yellow coloration is directly proportional to the amount of Human anti-antigen Ig captured on the plate.

      Answer

The Dengue IgG and IgM Kits have been validated for use on serum samples and plasma samples collected with citrate. Although it may well work we have not tried the kit with samples collected using EDTA. We would therefore not be able to guarantee the performance of the kit with these samples.

If you would like to purchase either ab108728 or ab108729 and use it with your serum prepared with EDTA, you would be eligible to participate in the Abtrial scheme. This offer means that if you purchase ab108728 or ab108729 as normal, test the kit with your samples in and let us know of the results through an Abreview (no matter whether positive or negative) you would then be eligible to claim the value of ab108728 or ab108729 off any product in our catalogue. More information on this offer can be found here:

      www.abcam.com/abtrial

Please note that the antibody to be tested must be purchased, tested, the Abreview submitted and the free product claimed within a 4 month period.

If you would be interested in participating in this scheme please do let me know as a discount code needs to be issued prior to the purchase of ab108728 or ab108729.

      Question

Customer kindly contacted us regarding the specificity of ab108728 for Dengue Virus serotypes 1-4. He was specifically wondering if we had any data showing that the antibody in the kit recognizes all 4 serotypes.

      Answer

I contacted the laboratory, and they confirmed that ab108728 does in fact cross react with Dengue Virus serotypes 1-4. It is very difficult to get samples with defined subtype of Dengue because the antibodies show a strong cross reactivity. Due to this cross reactivity it is sufficient to use Dengue-2 antigen (the antigen used in ab108728) to detect antibodies against subtypes 1-4. I have attached an extract of a WHO publication. On page 100 of the original document you will find the related statement in the paragraph right above the figure.

      Answer

This product is not in stock and currently has a 1 -2 week lead time. The kit will recognize all four serotypes (DEN-1, DEN-2, DEN-3, and DEN-4). While the Dengue Virus IgG ELISA is coated with purified Dengue 2 Virus proteins from strain 16681. Dengue Virus Type 2 is cross reactive with antibodies from Serotypes 1-4.

      Answer

The Dengue Virus IgG ELISA is coated with purified Dengue 2 Virus proteins from strain 16681. Dengue Virus Type 2 is cross reactive with antibodies from Serotypes 1-4, thus the kit will detect IgG from patient samples past infected with any of the four serotypes. The kit is not specific to Dengue Type 2.

It has not been tested for reactivity to west nile virus.

Might you be able to provide information on the specific anti-Dengue antibody used as capture antibody in the ELISA, particularly the immunogenic sequence (ie sequence injected into mouse or rabbit to produce the Ab?)

      Answer

In this assay, microtiter strip wells are precoated with Dengue Virus antigens type 2 to bind corresponding antibodies of the specimen. No capture antibody is used. The only antibody in the kit is the anti-human-HRP conjugate used to detect any antibody bound to the Dengue Virus antigens type 2.
