      Abcam’s Factor XIII Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of factor XIII in plasma, serum, milk, urine, saliva, and cell culture supernatants.


      A Factor XIII specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently a Factor XIII specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Complex is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of Factor XIII captured in plate.


      The entire kit may be stored at -20°C for long term storage before reconstitution - Avoid repeated freeze-thaw cycles.

      Factor XIII is activated by thrombin and calcium ion to a transglutaminase that catalyzes the formation of gamma-glutamyl-epsilon-lysine cross-links between fibrin chains, thus stabilizing the fibrin clot. Also cross-link alpha-2-plasmin inhibitor, or fibronectin, to the alpha chains of fibrin.

      Defects in F13A1 are the cause of factor XIII subunit A deficiency (FA13AD) [MIM:613225]. FA13AD is an autosomal recessive disorder characterized by a life-long bleeding tendency, impaired wound healing and spontaneous abortion in affected women.

      The kit using different antibodies, but the binding site is not mapped. Since they both polyclonal antibody, it may have multiple binding sites for protein.

      Question

      The OD's for the standard curve were very low- the highest standard measured at 0.21. Background was 0.046. Samples read around 0.1 (double the background). This seems very low, do you agree? After the streptavidin-peroxidase incubation, the plates were washed and left at room temperature for 45 min. Could this have caused the low signal? Do you have any suggestions to increase the signal?
      Also, I'm expecting that my plasma samples will have very low levels of the analyte. Can I dilute them less than 1:1000 (which the protocol instructs to do)? I would like to dilute some of my samples only 1:100, and others 1:300-1:500. Will this interfere with the assay? Will the higher concentration of chelating agent (EDTA) interfere?

      Thank you for your call last week and for your patience while I've been in touch with the lab regarding your questions.

      I've heard back from my contact at the lab, and she agrees that the OD values are lower than expected. An OD of 1.5-2.0 would be expected for the highest standard.She also believes this could have been caused by the breakduring the protocol, and she recommends repeating the experimentonce more without the 45 minute break, and compare the results.


      It is also fine to optimize the dilution of the samples, and 1:1000 is only a suggested startingdilution. it would be a good idea to test various dilutions to find the optimal dilution factor for your plasma samples.

      I am happy to confirm that the kit ab108836 against the human Factor XIII will recognize the full length protein as well as the fragment of the 37 first amino acids. I have just received the confirmation from the laboratory.

      I have checked also for the antibodies against Factor XIII:There is none which is specifically targeted to this region. We have several polyclonal antibodies which are against the full length protein. They might recognize the fragment as well, we can however not be sure. Please let me know if you would like me to investigate those further.
