    Abcam’s FGF9 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human FGF9 in plasma and cell culture supernatants. (Human FGF9 concentration is pretty low in normal plasma, it may not be detected in this assay).


    This assay employs an antibody specific for Human FGF9 coated on a 96-well plate. Standards and samples are pipetted into the wells and FGF9 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human FGF9 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of FGF9 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.


    We have not been able to detect the endogenous Human FGF9 in normal serum with ab100520, only in serum spiked with Human FGF9.

I have spoken to the lab and they have advised that you should not store the detection antibody concentrate for longer than 5 days at 4ºC. While not ideal, if you absolutely need to store it for a longer period of time, you should freeze it at -20ºC or -80ºC. Frozen it will likely be stable for 1-2 months.

The reason why we do not advise freezing the concentrate in the manual is to minimize the chances of degradation from freeze-thaw which can result in decreased signal strength. Therefore while it is best to keep the concentrate at 4ºC for just a few days, freezing the concentrate is a secondary option as long as only one freeze-thaw cycle occurs.

Currently the biotinylated detection antibody is not available from the catalogue; however I have requested that it be added, so that it can be purchased separately. It will take a few days for the product to be added. If you with to use ab84274, then we cannot guarantee that it will work.

Also, since you are using this kit on mouse species and we have only tested it on human speices, then the next time you purchase the kit, please let us know as you could qualify for our testing discount program:
