The Human Ficolin-2 Enzyme-Linked Immunosorbent Assay (ELISA) kit (ab213778) is designed for the quantitative measurement of Human Ficolin-2 in cell culture supernatants, serum and plasma (heparin).
The ELISA kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Ficolin-2 has been pre-coated onto 96-well plates. Standards (Expression system for standard: NSO; Immunogen sequence: L26-A313) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Ficolin-2 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human Ficolin-2 amount of sample captured in plate.

Notes
Ficolin-2, which was initially identified as L-ficolin, is a protein that in humans is encoded by the FCN2 gene. The product of this gene belongs to the ficolin family of proteins. This family is characterized by the presence of a leader peptide, a short N-terminal segment, followed by a collagen-like region, and a C-terminal fibrinogen-like domain. It is mapped to 9q34.3. This gene is predominantly expressed in the liver, and has been shown to have carbohydrate binding and opsonic activities. Serum concentration of ficolin-2 varies considerably in healthy individuals. FCN2 promoter polymorphisms were associated with marked changes in the ficolin-2 serum concentration.
