    Abcam’s GDNF (Glial Derived Neurotrophic Factor) Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme linked immunosorbent assay for the quantitative measurement of Human GDNF in plasma and cell culture supernatants. (Human GDNF concentration is pretty low in normal plasma, it may not be detected in this assay).

    This assay employs an antibody specific for Human GDNF coated on a 96-well plate. Standards and samples are pipetted into the wells and GDNF present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human GDNF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of GDNF bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

    We have not been able to detect the endogenous Human GDNF in normal serum with ab100525, only in serum spiked with Human GDNF.

The phenol red within the cell medium should not interfere with the measurement at 450nm but we would suggest running a media control just to make sure which can also act as your negative control.

With regards to the sample preparation, no additional pre-treatment of the customers samples is necessary when using cell supernatant however, it is true that acidifying the samples can enhance the activation of GDNF so if you wish to do this, you can but please remember to add alkali to neutralise the samples.

Thank you for contacting us. We do not currently sell any ELISA kits that are specifically designed for use on rat samples. After checking with the lab, our mouse and human ELISA kits for these targets will be unlikely to cross-react with rat, but have not been tested on rat samples.

I am happy to confirm that these kits should not be affected in their performanceif the packagewas not directly placed in therefrigeratorand thatthey have been left a day or two at room temperature.

I have also contacted the laboratory in this regards, and they confirmedas well that the kits are stable for afew days at room temperature:

"The ELISA kits can be stable for several days at room temperature based on our testing. Of course, we strongly recommend storing the kits at 2°C – 8°C upon receipt, but as you said the kit and its components should not have degraded from only one day unrefrigerated..."

Please doalso storethe standardsat -20°C after reconstitutionand avoid repeated freeze/thaw cycles.
