    ICA 512 / PTPRN in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of ICA 512 / PTPRN protein in Human  Citrate Plasma, Heparin Plasma, Cell and Tissue Extracts.


    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.


    Human ICA 512 / PTPRN plays a role in vesicle-mediated secretory processes (PubMed:24843546). Required for normal accumulation of secretory vesicles in hippocampus, pituitary and pancreatic islets. ICA 512 / PTPRN is required for the accumulation of normal levels of insulin-containing vesicles and preventing their degradation (PubMed:24843546) and plays a role in insulin secretion in response to glucose stimuli. Required for normal accumulation of the neurotransmitters norepinephrine, dopamine and serotonin in the brain. In females, but not in males, required for normal accumulation and secretion of pituitary hormones, such as luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Required to maintain normal levels of renin expression and renin release. ICA 512 / PTPRN appears to lack intrinsic enzyme activity but may regulate catalytic active protein-tyrosine phosphatases such as PTPRA through dimerization.

  • The concentrations of PTPRN were measured in duplicates, interpolated from the PTPRN standard curves and corrected for sample dilution. Undiluted samples are as follows: plasma (citrate) 25%, plasma (heparin) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PTPRN concentration was determined to be 874.85 pg/mL in plasma (citrate) and 819.33 pg/mL in plasma (heparin).

The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PTPRN concentration was determined to be 1478.61 pg/mL in SH-SY5Y extract and 489 pg/mL in human pancreas.
