  • Product overview

Abcam’s IFN gamma Human High Sensitivity in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of IFN gamma in Human sera, plasmas, buffered solutions and cell culture media.

A monoclonal antibody specific for IFN gamma has been coated onto the wells of the microtiter strips provided. Samples, including standards of known IFN gamma concentrations, control specimens or unknowns are pipetted into these wells. During the first incubation, the standards or samples and a biotinylated monoclonal antibody specific for IFN gamma are simultaneously incubated. After washing, the enzyme Streptavidin-HRP, that binds the biotinylated antibody is added, incubated and washed. A TMB substrate solution is added which acts on the bound enzyme to induce a colored reaction product. The intensity of this colored product is directly proportional to the concentration of IFN gamma present in the samples.

This kit will recognize both endogenous and recombinant Human IFN gamma.

Get results in 90 minutes with Human IFN gamma ELISA Kit (ab174443) from our SimpleStep ELISA® range.

  • Research areas

  • Function

Produced by lymphocytes activated by specific antigens or mitogens. IFN-gamma, in addition to having antiviral activity, has important immunoregulatory functions. It is a potent activator of macrophages, it has antiproliferative effects on transformed cells and it can potentiate the antiviral and antitumor effects of the type I interferons.
  • Tissue specificity

Released primarily from activated T lymphocytes.
  • Involvement in disease

In Caucasians, genetic variation in IFNG is associated with the risk of aplastic anemia (AA) [MIM:609135]. AA is a rare disease in which the reduction of the circulating blood cells results from damage to the stem cell pool in bone marrow. In most patients, the stem cell lesion is caused by an autoimmune attack. T-lymphocytes, activated by an endogenous or exogenous, and most often unknown antigenic stimulus, secrete cytokines, including IFN-gamma, which would in turn be able to suppress hematopoiesis.
  • Sequence similarities

Belongs to the type II (or gamma) interferon family.
  • Post-translational

Proteolytic processing produces C-terminal heterogeneity, with proteins ending alternatively at Gly-150, Met-157 or Gly-161.

Our Abpromise guarantee covers the use of ab46048 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
Sandwich ELISA Use at an assay dependent concentration.

  • IFN gamma measured in cell culture supernatants from control or treated (48 hours; 2% PHA-M; LifeTechnologies) PBMCs, diluted 1/100-1/10000 (duplicates +/- SD).

  • IFN gamma measured in undiluted cell culture supernatants from control or stimulated (48 hours with 50 ng x mL-1 PMA (ab120297) and 1 μM ionomycin (ab120116)) Jurkat cells. Control sample was below limit of detection (<0.57 pg x mL-1)  (duplicates +/- SD).

  • IFN gamma measured in undiluted human biological fluids; mouse and rat samples (plasma and serum) were below limit of detection (duplicates +/- SD).

  • Dilution curves of IFN gamma (ab46048) and NIBSC standard (87/586). One ng of standard IFN gamma corresponds to 12.3 (+/- 1.3) IU NIBSC 87/586. Background signal subtracted (duplicates; +/- SD).

Thank you very much for contacting us with your question.

We don't assign specific expiration dates for our products, but we expect the kit ab46048 to be viable for 12-18 months. I've looked at the specific lots on our most recent orders to your institution, and we would have removed these kits from our stock in June/July of next year. Our Abpromise guarantees that all products will work as stated on the datasheets for up to 6 months after purchase.

I hope that this information will be useful, but please let me know if you have any further questions and I'll be happy to help.

Thank you for contacting us with your questions, and for your patience while I have been in touch with the lab regarding your enquiries.
The lab scientists do not recommend storing the reconstituted standard because it is not stable and it may not work after storage. The wash buffer can be substitued with PBS with 0.05% Tween.

I hope this information will be useful, but please let me know if you have further questions or if there is anything else that we can do for you, and I'll be happy to help.

Thank you for contacting us.

I have confirmed that the standard for the Interferon Gamma Human ELISA Kit, ab46048, is not available as a stand-alone item.

The laboratory that developed the assay warns against storing the standard:

"We do not recommend to store any reconstituted standards, even at -20°C; our tests shows that the molecule is not stable at this concentration."

Please do not hesitate to contact us if you need any more advice or information.

Thank you for your patience. I have checked on the bovine IFN-gamma ELISA Kit. Unfortunately, the kit currently is not available in our catalog as the quality control tests have not yet been completed.  In general, due to the low homology between the human and bovine as well as guinea pig proteins, a human IFN gamma ELISA kit might most likely not work well for these two species, very unfortunately. I also checked our IFN gamma antibodies that were tested in ELISA, but unfortunately, we do not have any that were tested in bovine or guinea pig either. I am sorry to have no better news for you at this moment. Please do not hesitate to contact us if you need any more advice or information.

Thanks for your enquiry. I am very sorry to hear that you are having problems with this product. I have been discussing this enquiry with my colleagues in the Lab and would like to share the followings. 1) First, the regular volume of HRP diluent normally provided is 25.0ml and I will internally investigate how vials of only 23.0ml could have been dispatched. We can provide additional vial FOC for you if you wish. 2) Regarding low OD observed, the temperature can indeed be an explanation. The effect of lower temperature has not been fully studied and I have no data to show. However, generally based on physical-chemistry principles the temperature impacts the enzyme reactivity. In the High sensitivity kits configuration, enzyme is incubated twice (2 steps + the amplifier step) so it makes sense to consider high impact. 3) The main important factor for the good validation of the assay is the ratio between 0 and 25pg/ml standard point with 0.8 OD and less than 0.08 for zero, the assay remains fine. Hope this helps. Please do let me know how you wish to proceed with this enquiry.
