    IGFBP7 in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of IGFBP7 protein in human serum, plasma, urine, cell culture supernatants, and cell extracts.


    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.


    Insulin-like growth factor-binding protein 7, or IGFBP7 belongs to the IGFBP superfamily, which plays an integral role in regulating insulin-like growth factor (IGF) actions in a wide variety of cell types, including stimulation of prostacyclin production and cell adhesion. IGFBP7 is a low-affinity IGF binding protein. The protein functions through its binding to secreted growth factors including IGF1, insulin, and activin. IGFBP7 levels have been related to cancer development. Loss of expression of IGFBP7 has been associated with poor survival in multiple cancer types and with tumor chemotherapy resistance. IGFBP7 also has been identified as a cell cycle arrest biomarker for human acute kidney injury and serves as a prognostic indicator for early stage acute kidney injury development.

The mean IGFBP7 concentration was determined to be 205.0 ng/mL in neat serum, 196.4 ng/mL in neat plasma (citrate) and 189.9 ng/mL in neat plasma (EDTA).

The mean IGFBP7 concentration was determined to be 337.7 ng/mL in neat male urine, 397.6 ng/mL in neat female urine, and 389.5 ng/mL in neat HeLa cell culture supernatant.
