Target

  • Relevance

    IgG is a monomeric immunoglobulin, built of two heavy chains gamma and two light chains. Each molecule has two antigen binding sites. This is the most abundant immunoglobulin and is approximately equally distributed in blood and in tissue liquids, constituting 75% of serum immunoglobulins in humans. This is the only isotype that can pass through the human placenta, thereby providing protection to the fetus in its first weeks of life before its own immune system has developed. It can bind to many kinds of pathogens, for example viruses, bacteria, and fungi, and protects the body against them by complement activation (classic pathway), opsonization for phagocytosis and neutralisation of their toxins. There are 4 subclasses: IgG1 (66%), IgG2 (23%), IgG3 (7%) and IgG4 (4%).

    We tested ab109489 on mouse kidney, spleen, heart, brain, serum and thymus lysates in WB and did not see any bands at all (results attached). For this reason, we do not recommend the antibody for use with mouse nor do we expect cross-reactivity.

    I have compared the immunogen sequence similarity between human and Cynomolgus and have found 72.3% homology. We suggest that less than 85% homology is a good indicator that the antibody will not cross react however, we cannot guarentee this.

    Thank you for the prompt reply. The application would be in an immunoassay the methodology can be ECLIA, ELISA, SPR etc. The preference would be recognition of all IgG subclasses, however, the antibody should have been demonstrated to not react with (all) Cynomolgus IgG (subclasses).

    Unfortunately, we do not have any antibodies against huamn IgG which has been demonstrated to not react with (all) Cynomolgus IgG available in our catalog.

    We have one antibody ab109489 for which we have the immunogen for (https://www.abcam.com/ab109489). I have tried to do a sequence alignment to find out whether this antibody is likely to recognise Cynomolgus IgG however, I could not find the amino acid sequence for Cynomolgus IgG. If you have it, I would be happy to execute a sequence alignment for you.
