    IL-16 in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of IL-16 protein in human serum, plasma and cell culture supernatant samples.


    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

    IL-16 is a pro-inflammatory cytokine that stimulates a migratory response in CD4+ lymphocytes, monocytes, and eosinophils. IL-16 primes CD4+ T-cells for IL-2 and IL-15 responsiveness. It also induces T-lymphocyte expression of interleukin 2 receptor. IL-16 is synthesized in much larger 3 precursor isoforms localized in cytoplasm and nucleus and processed into 121 amino acid-long secreted IL-16. The isoform 1 may act as a scaffolding protein that anchors ion channels in the membrane. The isoform 3 is involved in cell cycle progression in T-cells. The isoform 3 appears to be involved in transcriptional regulation of SKP2 and is probably part of a transcriptional repression complex on the core promoter of the SKP2 gene. It may act as a scaffold for GABPB1 (the DNA-binding subunit the GABP transcription factor complex) and HDAC3 thus maintaining transcriptional repression and blocking cell cycle progression in resting T-cells.

  • Neat serum samples from 10 apparently healthy male donors (1M – 10M) were measured in triplicates using this kit. Interpolated data values are graphed in pg of IL-16 per mL of serum (mean +/- SD, n=3). The mean of IL-16 concentration of these serum samples was determined to be 75.7 pg/mL with a range of 48.7 – 122.9 pg/mL.

  • PBMC were grown in the absence or presence of phytohemagglutinin (PHA) for 48 hours. IL-16 concentrations were measured in neat and 2 X diluted cell culture supernatants of the unstimulated PBMC and the stimulated PBMC, and 10F RPMI1640 media. Raw data values (mean +/-SD, n=2) are graphed. The dotted line represents Blank control.
