      Abcam’s IL-8 Human in vitro ELISA Set is designed for the quantitative measurement of IL-8 in cell culture supernatants, buffered solutions, serum, plasma and other body fluids.


      The IL-8 ELISA Set includes both the capture and the detection antibodies as well as Horse-Radish peroxidase conjugated Streptavidin (HRP-Strep) and TMB which are the reagents used in the colour development step.


      This ELISA set is intended to measure human IL-8 in cell -culture supernatants, as well as in serum and plasma samples. The enclosed procedures provide general guidelines for ELISA preparation. This procedure may need some optimization depending on the plastic, matrix, standard range.

    • Function

      IL-8 is a chemotactic factor that attracts neutrophils, basophils, and T-cells, but not monocytes. It is also involved in neutrophil activation. It is released from several cell types in response to an inflammatory stimulus. IL-8(6-77) has a 5-10-fold higher activity on neutrophil activation, IL-8(5-77) has increased activity on neutrophil activation and IL-8(7-77) has a higher affinity to receptors CXCR1 and CXCR2 as compared to IL-8(1-77), respectively.

        Dear technical team,   Our customer purchased ab48481, ab47443 and ab48478.   She had experience using other bander's ELISA kit but ELIPAIR has different protocol that it needs 24hr drying time when coating the plate.   She thinks 2~3 hour is enough and wants to try short time drying so, please let her know why it needs 24 hour time to dry.   Second question is about reference filter. Is it necessary or optional?   "the absorbance at 630nm or 650nm" is different from other kit what she used before.   540~570nm are general absorbance for reference filter?   Please explain about this for our customer.   Best regards,

        Answer

        Thank you for contacting us. Overnight Incubation of plate at 4C is sort of general recommendation we provide to our customers. 3-4 hours at 4C can also be tried; the assay should be OK However in case of no readings we would be suggesting the overnight incubation also. The assay should read at 450nm wavelength which should be fine with standard filters. Any reference filter can be used. I hope this information is helpful to you.
